基于GC-MS的不同产地铁皮石斛代谢物差异分析 被引量:1

摘要 【目的】探究产地环境对铁皮石斛生长及代谢物质的成分表征影响,筛选不同产地的铁皮石斛差异代谢物。【方法】采用气相色谱质谱联用技术(GC-MS)结合主成分分析(PCA)、偏最小二乘判别分析(PLS-DA)等多元统计分析方法对不同产地铁皮石斛样品进行非靶向代谢组学分析。【结果】对不同产地石斛代谢物进行主成分分析可以将栽培品种与除湖南外的野生品种区分开,结合偏最小二乘判别分析与T检验(Student’s t-test)对湖南野生种与室内栽培种进行差异代谢物分析,并利用聚类热图分析(HCA)进行差异代谢物的验证,最终找到包括丙酸(Propionic acid)、苏氨酸(Threonine)等在内的20种极显著差异代谢物(P<0.01)。【结论】野生铁皮石斛与栽培品种间存在20种显著差异代谢物,主要为有机酸及多糖类代谢物,本研究可为指导铁皮石斛的植源性鉴定及真伪判断提供理论依据。 [Objective]The present paper aimed to explore the influence of the production area on the growth of Dendrobium candidum and the composition of metabolites,the different metabolites of D.candidum from different production areas were screened.[Method]The present study used gas chromatography-mass spectrometry(GC-MS)combined with principal component analysis(PC A),partial least square discriminant analysis(PLS-DA)and other multivariate statistical analysis methods for non-targeted metabolomics analysis of Dendrobium nobile samples from different productions.[Result]The principal component analysis of Dendrobium metabolites from different producing areas can distinguish cultivated varieties from wild varieties except Hunan,combined with partial least square discriminant analysis and T test(Student's-test)to compare Hunan wild species and indoor cultivated species to carry out differential metabolite analysis.The cluster heat map analysis(HCA)was used to verify the differential metabolites,and 20 extremely significant different metabolites were found finally including Propionic acid,Threonine(P<0.01).[Conclusion]There are 20 significantly different metabolites between wild D.candidum and cultivated varieties,mainly organic acid and polysaccharide metabolites.The study can provide a theoretical basis for the identification and authenticity of D.candidum.
