circRNA_0000285靶向miR-576-3p调控宫颈癌细胞增殖、迁移、侵袭的分子机制 被引量:1

circRNA_0000285 regulated the molecular mechanism of cervical cancer cell proliferation,migration and invasion by targeting miR-576-3p

摘要 目的探讨环状RNA_0000285(circularRNA_0000285,circRNA_0000285)对宫颈癌细胞生物学行为的影响及其作用机制。方法采用实时定量PCR(quantitative real-time reverse transcription PCR,qRT-PCR)法检测癌旁组织与宫颈癌组织中circRNA_0000285、微小RNA-576-3p(microRNA-576-3p,miR-576-3p)的表达水平;分别将si-NC、si-circRNA_0000285、miR-NC、miR-576-3pmimics、si-circRNA_0000285与anti-miR-NC、si-circRNA_0000285与anti-miR-576-3p转染入宫颈癌细胞HeLa;采用qRT-PCR法检测HeLa细胞中circRNA_0000285、miR-576-3p的表达水平;采用MTT实验、Transwell小室实验分别检测细胞增殖、迁移及侵袭能力;双荧光素酶报告实验检测circRNA_0000285与miR-576-3p的靶向关系。结果与癌旁组织比较,宫颈癌组织中circRNA_0000285的表达水平升高(P<0.05),miR-576-3p的表达水平降低(P<0.05);转染si-circRNA_0000285或转染miR-576-3pmimics可明显降低细胞活力(P<0.05),减少迁移及侵袭细胞数(P<0.05);双荧光素酶报告实验证实circRNA_0000285可靶向调控miR-576-3p;转染anti-miR-576-3p可明显逆转转染si-circRNA_0000285对HeLa细胞增殖、迁移及侵袭的作用。结论抑制circRNA_0000285表达可通过上调miR-576-3p的表达从而减弱宫颈癌细胞增殖、迁移及侵袭能力。 Objective To explore the effect of circRNA_0000285 on the biological behavior of cervical cancer cells and its mechanism.Methods The qRT-PCR method was used to detect the expression levels of circRNA_0000285 and miR-576-3p in adjacent tissues and cervical cancer tissues.si-NC,si-circRNA_0000285,miR-NC,miR-576-3p mimics,si-circRNA_0000285 and anti-miR-NC,si-circRNA_0000285 and anti-miR-576-3p were transfected into cervical cancer cell HeLa.qRT-PCR was used to detect the expression levels of circRNA_0000285 and miR-576-3p in HeLa cells.MTT experiment and Transwell chamber experiment were used to detect cell proliferation,migration and invasion ability respectively.The dual luciferase reporter experiment was used to detect the targeting relationship between circRNA_0000285 and miR-576-3p.Results Compared with adjacent tissues,the expression level of circRNA_0000285 in cervical cancer tissues was increased(P<0.05),and the expression level of miR-576-3p was decreased(P<0.05).Transfection of si-circRNA_0000285 or miR-576-3p mimics could significantly reduce the cell viability(P<0.05),and reduce the number of migration and invasion cells(P<0.05).The dual luciferase report experiment confirmed that circRNA_0000285 could target miR-576-3p.Transfection of anti-miR-576-3p could obviously reverse the effect of transfection of si-circRNA_0000285 on the proliferation,migration and invasion of HeLa cells.Conclusion Suppressing the expression of circRNA_0000285 could increase the expression of miR-576-3p to weaken the ability of cervical cancer cell proliferation,migration and invasion.
