circSLC8A1通过调控miR-301a-3p表达对卵巢癌SKOV3细胞增殖和凋亡的影响 被引量:1

Effect of circSLC8A1 on the proliferation and apoptosis of ovarian cancer SKOV3 cells by regulating the expression of miR-301a-3p

摘要 目的探讨circSLC8A1对卵巢癌SKOV3细胞增殖和凋亡的影响及分子机制。方法收集本院2016年1月至2020年12月37例卵巢癌患者的癌组织及癌旁组织,RT-qPC检测circSLC8A1和miR-301a-3p表达水平。卵巢癌细胞株SKOV3随机分为pcDNA-circSLC8A1组、pcDNA组、anti-miR-301a-3p组、anti-miR-NC组、pcDNA-circSLC8A1+miR-301a-3p组、pcDNA-circSLC8A1+miR-NC组。分别采用CCK-8法检测细胞活性;平板克隆实验检测细胞克隆形成数;流式细胞术检测细胞凋亡率;蛋白质印迹法检测蛋白表达;双荧光素酶报告实验检测circSLC8A1和miR-301a-3p的靶向关系。结果卵巢癌组织中circSLCA81表达水平低于癌旁组织,miR-301a-3p表达水平高于癌旁组织(P<0.05)。过表达circSLC8A1或抑制miR-301a-3p表达,细胞活性降低,细胞克隆形成数减少,细胞凋亡率及Cleaved-caspase3、Cleaved-caspase9蛋白表达水平升高(P<0.05)。circSLC8A1靶向负调控miR-301a-3p,上调miR-301a-3p表达逆转了circSLC8A1过表达对SKOV3细胞增殖和凋亡的作用。结论 circSLC8A1通过下调miR-301a-3p表达抑制卵巢癌SKOV3细胞增殖,促进其凋亡。 Objective To explore the effect of circSLC8 A1 on the proliferation and apoptosis of ovarian cancer SKOV3 cells and its molecular mechanism. Methods The cancer tissues and adjacent tissues of 37 ovarian cancer patients in our hospital from January 2016 to December 2020 were collected, RT-qPCR was used to detect the expression levels of circSLC8 A1 and miR-301 a-3 p;ovarian cancer cells Strain SKOV3 was randomly divided into pcDNA-circSLC8 A1 group,pcDNA group, anti-miR-301 a-3 p group, anti-miR-NC group, pcDNA-circSLC8 A1+miR-301 a-3 p group, pcDNA-circSLC8 A1+miR-NC group;cells Counting kit 8(CCK-8) to detect cell viability;plate cloning experiment to detect the number of cell clones;flow cytometry to detect cell apoptosis rate;Western blot to detect protein expression;dual luciferase report experiment to detect the targeting relationship between circSLC8 A1 and miR-301 a-3 p. Results The expression level of circSLCA81 in ovarian cancer tissue was lower than that in adjacent tissues, and the expression level of miR-301 a-3 p was higher than that in adjacent tissues(P<0.05). Overexpression of circSLC8 A1 or inhibition of miR-301 a-3 p expression, cell viability was decreased, the number of cell clones formed was decreased, the cell apoptosis rate, the protein expression levels of Cleaved-caspase3 and Cleaved-caspase9 were increased(P<0.05). circSLC8 A1 targeted and negatively regulated miR-301 a-3 p;up-regulating the expression of miR-301 a-3 p reversed the effect of circSLC8 A1 overexpression on the proliferation and apoptosis of SKOV3 cells. Conclusions circSLC8 A1 can inhibit the proliferation of ovarian cancer SKOV3 cells and promotes their apoptosis by down-regulating the expression of miR-301 a-3 p.
