生物节律蛋白、乙醇脱氢酶4、下丘脑泌素受体2基因多态性与丛集性头痛关联性的Meta分析

摘要 目的探讨生物节律蛋白(CLOCK)、乙醇脱氢酶4(ADH4)、下丘脑泌素受体2(HCRTR2)基因多态性和丛集性头痛(CH)的关系。方法系统检索PubMed、Cochrane Library、EMBASE、Web of Science、中国知网和万方数据库。根据纳入和排除标准筛选出符合条件的研究,借助纽卡斯尔-渥太华量表(NOS)评价纳入文献的质量。利用RevMan 5.3软件计算出不同遗传模型下的OR和95%CI,并对统计结果进行敏感性分析。结果共纳入13篇文献,包括15项独立研究,均为高质量研究。Meta分析结果显示,CLOCK rs1801260、ADH4 rs1800759和rs1126671,HCRTR2 rs2653349、rs3122156、rs2653342多态性位点均与CH无关(均P>0.05)。敏感性分析确认了Meta分析结果的稳定性和可靠性。结论CLOCK rs1801260、ADH4 rs1800759和rs1126671,HCRTR2 rs2653349、rs3122156、rs2653342多态性和CH无关。考虑到本研究的局限性,仍有必要在不同种族人群中开展大样本研究来进一步验证本研究的结果。 Objective To explore the association of circadian locomotor output cycles kaput(CLOCK),alcohol dehydrogenase 4(ADH4),hypocretin receptor type 2(HCRTR2)genes’polymorphisms and cluster headache(CH).Methods PubMed,Cochrane Library,EMBASE,Web of Science,CNKI and Wanfang database were systematically searched.Eligible studies were screened according to the inclusion and exclusion criteria,and the quality of the included literature was evaluated by the Newcastle-Ottawa scale(NOS).The OR and 95%CI under different genetic models were calculated by RevMan 5.3 software,and the sensitivity analysis of the statistical results was performed.Results A total of 13 papers were included,including 15 independent studies,which were all high-quality studies.The results of the meta-analysis showed that there was no relationship between the polymorphisms of CLOCK rs1801260,ADH4 rs1800759 and rs1126671,HCRTR2 rs2653349,rs3122156 and rs2653342 and CH(all P>0.05).Sensitivity analysis confirmed the stability and reliability of meta-analysis results.Conclusions There is no relationship between the polymorphisms of CLOCK rs1801260,ADH4 rs1800759 and rs1126671,HCRTR2 rs2653349,rs3122156 and rs2653342 and CH.Considering the limitations of this study,it is still necessary to carry out large sample studies in different ethnic groups to further validate the results of this study.
