摘要
为优化厚垣孢普可尼亚菌(Pochonia chlamydosporia)菌株PC7的液态发酵条件,研究不同发酵条件下厚垣孢普可尼亚菌PC7的产孢量与菌丝生物量,分析培养温度、初始pH、供氧量、接菌量对于PC7菌株液态发酵的影响。结果表明:PC7菌株培养液在培养温度为26℃、初始pH值为8、500 mL三角瓶装瓶量200 mL时,以0.5 mL为初始接菌量,产孢量在培养5~7 d时达到最大,是最适产孢条件。以菌丝生产为目标,进行液体发酵生产时,温度24℃、初始pH值为8、500 mL三角瓶装瓶量150 mL时,以1.0 mL为初始接菌量,菌丝干质量在培养5 d时达到最大,有利于提高菌丝产量,是最适培养条件。
To optimize the liquid fermentation conditions of Pochonia chlamydosporia PC7 strain,we studied the sporulation quantity and hyphae biomass of PC7 under different fermentation conditions by analyzing the effects of incubation temperature,initial pH of culture medium,oxygen supply and inoculation amount on the liquid fermentation of PC7 strain,in order to supply technical support for the future large-scale industrial production.The incubation temperature of PC7 strain is incubated at 26℃,initial pH value is 8,200 mL bottle volume(triangle bottle capacity was 500 mL),0.5 mL as the initial inoculation amount,and the sporulation quantity reaches the maximum after 5-7 d of culture,which is the optimal condition for sporulation.With mycelium production as the target,when liquid fermentation production is carried out,the incubation temperature is 24℃,the initial pH value is 8,the bottling volume of 150 mL(triangular bottle capacity was 500 mL)and 1.0 mL as the initial inoculation amount,the dry weight of mycelium reaches the maximum after 5-d culture,which is conducive to improving the yield of mycelium,and is the optimal incubation condition.
