不同生境人参茎叶总皂苷对心律失常小鼠的作用比较

摘要 目的探讨园参茎叶总皂苷(GSLS)和林下山参茎叶总皂苷(MSLS)对心律失常小鼠的改善作用。方法将80只SPF级BALB/c小鼠随机分为正常组、模型组、园参茎叶总皂苷低(GSLS-L)、中(GSLS-M)、高(GSLS-H)剂量组、林下山参茎叶总皂苷低(MSLS-L)、中(MSLS-M)、高(MSLS-H)剂量组。各给药组小鼠预先给予相应药物7 d。末次给药1.5 h后,采用尾静脉注射0.4%氯化钡溶液制备小鼠心律失常模型,并通过生物信号采集处理系统监测模型组和给药组小鼠Ⅱ导联心电图;采用ATP酶测试盒检测各组小鼠心肌组织中Na^(+)-K^(+)-ATP酶和Ca^(2+)-Mg^(2+)-ATP酶的表达。结果与模型组相比,园参茎叶总皂苷低、中、高剂量组和林下山参茎叶总皂苷低、中、高剂量组心律失常持续时间均有所降低(P<0.01),园参茎叶总皂苷和林下山参茎叶总皂苷各同等剂量组比较未见统计学差异;园参茎叶总皂苷和林下山参茎叶总皂苷对模型小鼠心电图的P波、QRS波和PQ间期均具有明显改善作用(P<0.01,P<0.05);园参茎叶总皂苷中、高剂量组和林下山参茎叶总皂苷中、高剂量组小鼠心肌组织中Na^(+)-K^(+)-ATP酶及Ca^(2+)-Mg^(2+)-ATP酶表达水平显著升高(P<0.01),园参茎叶总皂苷和林下山参茎叶总皂苷各同等剂量组比较未见统计学差异。结论园参茎叶总皂苷和林下山参茎叶总皂苷均可以通过升高氯化钡致心律失常小鼠心肌组织中Na^(+)-K^(+)-ATP酶及Ca^(2+)-Mg^(2+)-ATP酶的表达来改善心律失常,并且各同等剂量组间未见统计学差异。 Objective To explore the efficacy of garden ginseng stem-leaf saponins(GSLS)and mountain cultivated ginseng stem-leaf saponins(MSLS)on mice with arrhythmia.Methods Eighty SPF BALB/c mice were randomly divided into normal group,model group,GSLS low(GSLS-L),medium(GSLS-M),high(GSLS-H)dose groups,MSLS low(MSLS-L),medium(MSLS-M),high(MSLS-H)dose groups.Mice in administration group were given corresponding drugs in advance for 7 days.1.5 h after the last administration,mice with arrhythmia was established by tail vein injection of 0.4%barium chloride solution.TheⅡ-lead electrocardiogram of mice in model group and administration group were monitored by biological signal acquisition system.The expression of Na^(+)-K^(+)-ATPase and Ca^(2+)-Mg^(2+)-ATPase in myocardial tissues was detected by ELISA kits.Results Compared with model group,the duration of arrhythmia in GSLS-L,GSLS-M,GSLS-H groups and MSLS-L,MSLS-M,MSLS-H groups were decreased(P<0.01),and there was no statistical difference between GSLS and MSLS at the same dose;In addition,GSLS and MSLS significantly improved the P wave,QRS wave and PQ interval of model mice(P<0.01,P<0.05);The expression of Na^(+)-K^(+)-ATPase and Ca^(2+)-Mg^(2+)-ATPase in myocardial tissue of mice in GSLS-M,GSLS-H,MSLS-M and MSLS-H groups were significantly increased(P<0.01),and there was no statistical difference between GSLS and MSLS at the same dose.Conclusion GSLS and MSLS can improve arrhythmia by increasing the expression of Na^(+)-K^(+)-ATPase and Ca^(2+)-Mg^(2+)-ATPase in myocardial tissue,and there was no statistical difference between GSLS and MSLS at the same dose.
