摘要
细胞受到外界刺激导致DNA损伤时,DNA修复蛋白被激活并发挥功能,受损DNA得到修复。近年来研究表明,转录因子特异性蛋白1(specificity protein 1,Sp1)在DNA损伤修复中起重要作用,是参与调控DNA损伤应答与修复的重要蛋白之一。Sp1在多种肿瘤中高表达,通过调节细胞增殖、迁移和侵袭促进肿瘤的发生和发展,不利于肿瘤治疗。本文首先介绍Sp1特有的结构及其生理功能,由此引入Sp1通过转录依赖方式调控DNA损伤应答,其次介绍Sp1通过非转录依赖方式参与DSB修复,以及Sp1在肿瘤治疗中的研究进展,最后展望研究Sp1的意义及可能具有的应用前景。
When a cell is damaged by external stimuli,the DNA repair proteins are activated and function,and the damaged DNA is repaired.In recent years,it has been shown that transcription factor specific protein 1(Sp1)plays an important role in DNA damage repair and is one of the important proteins involved in the regulation of DNA damage response and repair.Sp1 is highly expressed in a variety of tumors,which promotes the occurrence and development of tumor by regulating cell proliferation,migration and invasion.In this paper,the specific structure and physiological functions of Sp1 were first introduced to clarify the regulation of DNA damage repair by Sp1 in a transcriptional-dependence way,followed by the non-transcription-dependent involvement of Sp1 in DSB repair,as well as the research progress of Sp1 in tumor.Finally,the significance of Sp1 and its possible application prospects research are prospected.
