响应面优化流苏石斛多糖提取工艺 被引量:5

Optimization of Extraction Technique of Polysaccharides of Dendrobium fimbriatum by Response Surface Methodology

摘要 以流苏石斛为原料,采用超声辅助技术研究液料比、提取时间、提取温度对流苏石斛中多糖的影响。在单因素试验基础上,利用响应面分析法中Box-Behnken中心组合设计原理分析优化流苏石斛中多糖的提取工艺。结果表明, 3个因素的影响大小为提取时间>提取温度>液料比,最佳提取工艺条件为液料比50.445∶1 mL/g、提取时间40.744 min、提取温度60.893℃,在此条件下,预测多糖得率10.329%,实际值为10.19%,二者误差在1.5%以内。试验表明该回归模型合理有效,研究所得到的优化工艺具有一定可行性,可为流苏石斛多糖的提取提供参考,具有广阔应用前景。 To optimize the extraction technology for polysaccharides from the Dendrobium fimbriatum, using Dendrobium fimbriatum as the raw material, the polysaccharide was extracted by ultrasonic assisted technique. On the basis of single factor experiment, the design principle of Box-Behnken center combination in response surface methodology(RSM) was applied to analyze and optimize the technological conditions such as liquid-material ratio, extraction time and extraction temperature.The size of three factors was extraction time>extraction temperature>liquid-material ratio, the optimal extraction condition was liquid material ratio 50.445∶1 mL/g, extraction time 40.744 min and extraction temperature of 60.893 ℃, the rate of the actual value was 10.19% polysaccharides under this condition, and the theoretical value was 10.329%. The error of the two was within 1.5%. The regression model was reasonable and effective, and the optimized process obtained in the research had a certain feasibility. It could provide a reference for the extraction of Dendrobium fringe polysaccharides and had broad application prospects.
