摘要
建立HPLC法同时测定荞麦中8种酚酸类物质的方法。该方法采用高效液相色谱法,选用Agilent Zorbax SB-C18色谱柱(4.6 mm×250 mm,5μm),柱温25℃;流动相为0.1%甲酸水溶液-乙腈-甲醇,梯度洗脱;流速0.8 mL/min;进样量20μL;基质加标-外标法定量。结果表明,在2.0~20μg/mL质量浓度范围内,其浓度和峰面积有良好的线性关系,相关系数R≥0.99926。样品加标回收率在71.2%~97.36%之间,S_(RSD)<7.64%(n=6)。该方法前处理简单、准确、重现性好,适用于荞麦中酚酸类成分的定量分析。
To establish a high performance liquid chromatography(HPLC)method for simultaneous determination of 8 phenolic acids in buckwheat,the method was established by HPLC with Agilent Zorbax SB-C18 column(4.6 mm×250 mm,5μm),column temperature of 25℃and mobile phase of 0.1%formic acid aqueous-acetonitrile-methanol,gradient elution.Flow rate was 0.8 mL/min,injection was 20μL,and matrix addition external standard method was used.The results showed that there was a good linear relationship between the concentration and the peak area in the concentration range of 2.0-20μg/m L,and the correlation coefficient R≥0.99926.The recoveries were 71.2%-97.36%,and S_(RSD)<7.64%(n=6).The method was simple,accurate and reproducible,which was suitable for the quantitative analysis of phenolic acids in buckwheat.
