摘要
试验旨在建立婴幼儿配方乳粉中2’-岩藻糖基乳糖(2’-FL)的定量检测方法。用磺基水杨酸溶液作蛋白质沉淀剂,利用滤膜和净化柱对样品进行净化,通过高效阴离子交换色谱-脉冲电化学检测进行测定。结果显示,该方法在0.223 2~17.856μg/mL范围内线性关系良好,相关系数R2为0.999 4;该方法对乳粉基质定量限为100 mg/100 g;加标量在200~400 mg/100 g时,加标回收率在93.98%~99.35%,相对标准偏差(S_(RSD))为0.30%~1.65%。该方法检测结果准确可靠,适用于婴幼儿配方乳粉中2’-FL的定量检测。
This study aimes to establish a quantitative method for the determination of 2’-fucosyllactose(2’-FL) in infant formula. Sulfosalicylic acid solution was used as protein precipitator, and the samples were purified by filtration membrane and purification column. The samples were determined by high perfomance anion exchange chromatography with pulsed electrochemical detection(HPAEC-PED). The results showed that this method showed good linear responses ranging from 0.223 2-17.856 μg/mL with satisfactory linear correlation coefficient of 0.999 4. The limit of quantification(LOQ) was 100 mg/100 g in milk power matrix. When the scalar was 200-400 mg/100 g, the recoveries were 93.98%-99.35%, and the relative standard deviation(S_(RSD)) was 0.30%-1.65%. The technique exhibited high reliability and accuracy, which was suitable for the detection of infant formula samples.
