碱性成纤维细胞因子联合维生素E对色素沉着模型的实验研究 被引量:1

Experimental study on basic fibroblast growth factor combined with vitamin E for guinea pig model of pigmentation

摘要 目的探究水光注射碱性成纤维细胞因子(basic fibroblast growth factor,bFGF)联合外用维生素E对豚鼠色素沉着作用的影响。方法2021年5—8月,河北工程大学临床医学院将15只雌性豚鼠311 nm UVB(中波紫外线)照射,建立色素沉着模型,随机分为3组。A组(5只):水光注射bFGF+外用维生素E。B组(5只):水光注射透明质酸+外用维生素E。C组(5只):水光注射平衡盐溶液。3组分别在注射前、注射后2周进行皮肤宏观学检测、Masson-Fontana法黑色素染色和ELisa试剂盒检测。结果A、B两组注射2周后,皮肤宏观学得分较注射前明显降低,差异有统计学意义(P<0.05);C组注射前后皮肤宏观学得分差异无统计学意义(P>0.05)。A、B两组注射2周后较注射前黑色素颗粒数量明显减少,差异有统计学意义(P<0.05);且注射2周后A组黑色素数量较B组明显减少,差异有统计学意义(P<0.05);C组注射前后差异无统计学意义(P>0.05)。A组黑素皮质素-1受体(melanocortin 1 receptor,MC1R)水平在注射后2周明显下降,差异有统计学意义(P<0.05)。结论bFGF联合外用维生素E有消除皮肤黑色素、改善肤质、治疗光老化的作用。 Objective To explore the efficacy of basic fibroblast growth factor(bFGF)combined with vitamin E injection for guinea pig model of pigmentation.Methods Fifteen female guinea pigs were selected and back skin was shaved and irradiated by 311 nm UVB(medium wave ultraviolet)to establish pigmentation model.All female guinea pigs were randomly divided into 3 groups,5 cases in each group.The bFGF injection combined with external application of vitamin E was performed in group A.The hyaluronic acid injection combined with external application of vitamin E was performed in group B.The balanced salt solution injection was performed in group C.The skin macroscopic examination,Masson-Fontana staining for melanin granules and ELISA test kit was performed at immediately before injection and 2 weeks after injection.Results The macroscopic skin scores of group A and B at 2 weeks after injection were significantly lower than that before injection,the difference was statistically significant(P<0.05).The difference in macroscopic skin scores of group C before and after injection was not statistically significant(P>0.05).The number of melanin granules in group A and B was significantly reduced at 2 weeks after injection compared with that before injection,the difference was statistically significant(P<0.05).The number of melanin granules in group A was significantly reduced at 2 weeks after injection compared with that in group B,the difference was statistically significant(P<0.05).The difference was not statistically significant in group C before and after injection(P>0.05).The level of melanocortin-1 receptor(MC1R)in group A decreased significantly at 2 weeks after injection,the difference was statistically significant(P<0.05).Conclusion The bFGF combined with external application of vitamin E can eliminate skin melanin and improve skin quality with definite curative effect.
