摘要
为探究KIAA2012和UTP20多态性与胸椎数间的关联性,筛选胸椎数相关重要分子标记,利用Sequenom MassARRAY^(■)SNP技术检测苏尼特羊和小尾寒羊中与胸椎数相关候选基因KIAA2012和UTP20的单核苷酸多态性(SNPs)位点,开展群体遗传学分析,并与胸椎数进行关联分析;利用生物信息学分析工具对KIAA2012和UTP20的遗传特征进行分析。结果表明,UTP20 g.170058254G>A位点在苏尼特羊群体中突变型(GA)胸椎数显著(P<0.05)高于野生型(GG),g.170053888G>A和g.170072471G>A在小尾寒羊和苏尼特羊中均与胸椎数没有显著相关;KIAA2012 g.203440537A>G、g.203424302A>G、g.203440384A>T在小尾寒羊和苏尼特羊中与胸椎数均没有显著相关(P>0.05)。SNPs与胴体长度分析结果表明,在小尾寒羊KIAA2012 g.203440537A>G中,突变型(GA)胴体长度显著(P<0.05)高于野生型(GG)。生物信息学分析表明,KIAA2012为亲水蛋白,UTP20为疏水蛋白,两者蛋白结构均不稳定,无跨膜结构域。综上所述,UTP20 g.170058254G>A位点与苏尼特羊胸椎数性状显著相关,KIAA2012 g.203440537A>G与小尾寒羊胴体长度性状显著相关,可作为调控绵羊胸椎数变异潜在的分子辅助标记。
This study was to explore the relationship between the thoracic vertebrae number and polymorphisms of the KIAA2012 and UTP20 genes,and to screen the important molecular markers related to the thoracic vertebrae number.The single nucleotide polymorphisms(SNPs)sites related to the thoracic vertebrae number of KIAA2012 and UTP20 in Sunite sheep and Small Tail Han sheep was typed using the Sequenom MassARRAY^(■)SNP technology,and population genetic and correlation analysis associated with the thoracic vertebrae number were carried out.Bioinformatics analysis tools were used to analyze the genetic characteristics of KIAA2012 and UTP20.The results showed that the thoracic vertebrae number of the mutation type(GA)in the Sunite sheep population at the g.170058254 G>A site of the UTP20 gene was significantly higher(P<0.05)than that in the wild type(GG),and g.170053888 G>A and g.170072471 G>A were not significantly associated with the thoracic vertebrae number in the Small Tail Han sheep and Sunite sheep(P>0.05).All the three SNPs of KIAA2012(g.203440537 A>G,g.203424302 A>G,and g.203440384 A>T)were not significantly correlated with the thoracic vertebrae number in the Small Tail Han sheep and Sunite sheep(P>0.05).The correlation analysis between the SNPs and carcass length of the tested sheep showed that the mutation type(GA)in g.203440537A>G of the K1AA20X2 in the Small Tail Han sheep was significantly(P<0.05)higher than that in the wild type(GG).Bioinformatics analysis showed that KIAA2012 is a hydrophilic protein,while UTP20 is a hydrophobin.The structures of the two proteins were unstable,which had no transmembrane domains and might not have participated in the transmembrane biochemical reactions.Taken together,the g.170058254G>A locus of the UTP20 gene was significantly related to the thoracic vertebral number traits in the Sunite sheep,and the g.203440537A>G locus of the A7A42012 gene was significantly related to the carcass length traits in the Small Tail Han sheep.The two SNPs were expected to be used as potential molecular markers to regulate changes in the number of thoracic vertebrae in sheep.
