没食子酸对过敏性鼻炎小鼠的抗过敏作用研究 被引量:4

摘要 目的探究没食子酸对过敏性鼻炎(AR)小鼠的抗过敏作用。方法将100只小鼠随机分为空白组、模型组,低剂量实验组、中剂量实验组和高剂量实验组,每组20只。空白组小鼠不做处理,其他组小鼠通过卵清蛋白(OVA)诱导构建AR模型,低、中、高剂量实验组中分别采用20,40和80 mg·kg^(-1)没食子酸进行治疗。用酶联免疫吸附法检测炎症因子、OVA特异性免疫球蛋白水平;用流式细胞术检测嗜酸性粒细胞水平;用苏木精-伊红染色法染色观察小鼠鼻部病理情况并测定其厚度。结果空白组、模型组、低、中和高剂量实验组小鼠外周血γ干扰素(IFN-γ)水平分别为(13.47±0.86),(5.21±0.26),(9.67±3.14),(12.35±1.64)和(14.39±1.37)pg·mL^(-1);这5组的卵清蛋白特异性(IgE)分别为(1.37±0.31),(15.32±0.67),(14.62±2.38),(11.38±2.36)和(7.61±0.72)pg·mL^(-1);这5组的嗜酸性粒细胞分别为5.39±1.28,32.57±10.38,8.25±2.67,6.31±2.94和4.95±1.43;这5组的鼻粘膜厚度分别为(11.24±0.34),(24.35±3.67),(20.34±1.57),(17.34±2.34)和(14.86±1.37)μm。上述指标,低、中、高剂量实验组与模型组比较,差异均有统计学意义(均P<0.05)。结论没食子酸可以显著缓解AR小鼠的过敏反应。 Objective To investigate the anti-allergic effect of gallic acid on mice with allergic rhinitis.Methods One hundred mice were randomly divided into blank group,control group,experimental-L group,experimental-M group and experimental-H group,20 mice in each group.Mice in the blank group were left untreated,and mice in the other groups were induced to construct allergic rhinitis by ovalbumin(OVA),and 20,40 and 80 mg·kg^(-1) gallic acid were used in the experimental-L group,experimental-M group and experimental-H group,respectively.The levels of inflammatory factors,OVA-specific immunoglobulins were measured by enzyme linked immunosorbent assay.Eosinophil levels were detected by flow cytometry.The nasal pathology of mice was observed by hematoxylin-eosin staining and its thickness was detected.Results Peripheral blood gamma interferon (IFN-γ) levels in mice in the blank,control,experimental-L,experimental-M and experimental-H group were (13.47±0.86),(5.21±0.26),(9.67±3.14),(12.35±1.64) and (14.39±1.37) pg·m L^(-1);ovalbumin specific (IgE) was (1.37±0.31),(15.32±0.67),(14.62±2.38),(11.38±2.36) and (7.61±0.72) pg·m L^(-1);eosinophils were 5.39±1.28,32.57±10.38,8.25±2.67,6.31±2.94 and 4.95±1.43;nasal mucosal thickness were (11.24±0.34),(24.35±3.67),(20.34±1.57),(17.34±2.34) and (14.86±1.37)μm,with significant differences between the experimentalL,experimental-M,experimental-H group and the control group (P<0.05).Conclusion Gallic acid can significantly alleviate allergic reactions in mice with allergic rhinitis.
