云实种子的卡山烷二萜类化学成分及其生物活性研究 被引量:1

Study on the Chemical Constituents and Their Biological Activities of Cassane-type Diterpenes from Seeds of Caesalpinia Decapetala

摘要 目的:研究云实种子的化学成分并开展生物活性研究。方法:采用硅胶柱、Sephadex LH-20等柱色谱方法对云实种子95%甲醇提取物进行分离纯化,结合波谱技术与化学方法进行结构鉴定;采用MTT法进行抗肿瘤活性筛选,运用LPS诱导的RAW 264.7细胞释放NO的方法进行抗炎活性筛选。结果:从云实种子95%甲醇提取物中分离得到了13个化合物,结构鉴定为α-caesalpin(1)、 Neocaesalpin L(2)、 bonducellpin C acetate(3)、bonducellpin A(4)、 Caesalmin E(5)、Caesalmin C(6)、 Neocaaesalpin MP(7)、 1-deacteoxy-1-oxocaesalmin C(8)、Neocaesalpin AA(9)、 bonducellpin C(10)、 bonducellpin E(11)、Neocaaesalpin N(12)、 Caesalmin F(13)。体外抗肿瘤活性筛选发现化合物9对人宫颈癌细胞HeLa具有较强的抑制作用,IC;为26.5μmol·L;LPS诱导的RAW 264.7抗炎活性筛选表明化合物7和12具有一定的抗炎活性,IC;分别为31.6和38.2μmol·L;。结论:化合物2~11为首次从云实种子中分离得到;初步实验显示化合物9有较强的肿瘤抑制作用,而化合物7和12具有一定抗炎活性。 Objective: To study the chemical constituents and biological activities of cassane-type diterpenes from the seeds of Caesalpinia decapetala. Methods: The constituents were isolated and purified by silica gel chromatography and Sephadex LH-20 repeatedly, and the structures were identified by spectral analyses. MTT assay was used to explore the anti-tumor activity, and LPS induced NO release from RAW 264.7 cells was used to study the anti-inflammatory activity. Results: Thirteen compounds were isolated from the seeds of C. decapetala and the structures were identified as α-caesalpin(1), neocaesalpin L(2), bonducellpin C acetate(3), bonducellpin A(4), caesalmin E(5), caesalmin C(6), neocaaesalpin MP(7), 1-deacteoxy-1-oxocaesalmin C(8), neocaesalpin AA(9), bonducellpin C(10), bonducellpin E(11), neocaaesalpin N(12) and caesalmin F(13). The anti-tumor activity test in vitro displayed that the IC;of compound 9 was 26.5μmol·L;the anti-inflammatory activity in Raw 264.7 induced by LPS showed that compounds 7 and 12 had certain anti-inflammatory activity with the IC;of 31.6 and 38.2 μmol·L;, respectively. Conclusion: Compounds 2-11 were obtained from seeds of C. decapetala for the first time, and preliminary experiments showed that compound 9 displayed significant anti-tumoractivity, compounds 7 and 12 had moderate anti-inflammatory activities.
