The effect of different inactivation methods on the efficiency of nucleic acid detection of SARS-CoV-2 samples

摘要 目的研究2种灭活方法对新型冠状病毒样本核酸检测效率的影响。方法采用荧光定量PCR检测方法,比较和评价2种灭活方法对新型冠状病毒标本核酸检测的影响。结果56℃灭活后的标本核酸检测CT值有所增加,弱阳性标本出现假阴性结果。胍盐保存液灭活对标本核酸检测结果无负面影响。结论56℃灭活会降低新型冠状病毒标本核酸检测效率,建议用胍盐保存液进行检测前标本灭活处理。 Objective To study the effect of two inactivation methods on the efficiency of nucleic acid detection of SARS-CoV-2 samples.Methods The fluorescence quantitative PCR detection method was used to compare and evaluate the differences between two inactivation methods for the nucleic acid detection of SARS-CoV-2 samples.Results The CT value of nucleic acid detection increased after inactivation of the samples at 56℃,and the weakly positive samples showed false negative results.There was no negative effect on the nucleic acid test results of the samples after the inactivation with the guanidine salt preservation solution.Conclusion Inactivation at 56℃may reduce the efficiency of nucleic acid detection of SARS-CoV-2 samples,and inactivation with guanidine salt preservation solution is recommended which does not affect the efficiency of nucleic acid detection of SARS-CoV-2 samples.
