整合素α5在子宫内膜癌组织中的表达及其对子宫内膜癌细胞恶性生物学行为的影响 被引量:1

摘要 目的探讨整合素α5(ITGα5)在子宫内膜癌组织中的表达及其与患者临床病理特征的关系,以及ITGα5对子宫内膜癌细胞增殖、迁移与侵袭的影响。方法收集2019年2月至2020年12月于新疆医科大学第二附属医院行手术治疗的90例子宫内膜癌患者的肿瘤组织和癌旁组织标本,以及培养人子宫内膜癌细胞株HEC-1B作为研究对象。采用免疫组织化学法检测肿瘤组织和癌旁组织中ITGα5的表达,分析其与临床病理参数的关系;根据HEC-1B细胞中转染物质的不同,将其分为Control组(未进行任何处理)、NC组(转染si-NC)、ITGα5组(转染siRNA-ITGα5),采用Western blot法检测转染前后各组ITGα5蛋白表达水平,采用实时荧光定量聚合酶链反应(qRT-PCR)检测ITGα5-mRNA的表达水平;采用MTT实验检测细胞增殖能力,流式细胞术检测细胞周期,Transwell小室实验检测细胞侵袭能力,划痕实验检测细胞迁移能力。结果子宫内膜癌组织中ITGα5阳性率(58.89%)高于癌旁组织(13.33%),差异具有统计学意义(P=0.000);ITGα5阳性率与病理类型、病理分期、分化程度、淋巴结转移有关(P=0.000);沉默ITGα5后HEC-1B细胞中ITGα5-mRNA、ITGα5蛋白表达水平降低(P<0.05),HEC-1B细胞的增殖、迁移和侵袭能力均显著降低(P<0.01)。结论ITGα5在子宫内膜癌组织中高表达,沉默ITGα5可抑制HEC-1B细胞的增殖、迁移和侵袭,其在子宫内膜癌的发生发展中起重要作用。 Objective To investigate the expression of integrinα5(ITGα5)in endometrial carcinoma and its relationship with clinicopathological features,and the effect of interfering with ITGα5 on the proliferation,migration and invasion of endometrial carcinoma cell.Methods Tumor tissue and adjacent tissue specimens and human endometrial carcinoma cell line HEC-1B were collected from 90 cases of patients with endometrial carcinoma who underwent surgical treatment in the Second Affiliated Hospital of Xinjiang Medical University from February 2019 to December 2020.Immunohistochemistry was used to detect the expression of ITGα5 in tumor tissues and adjacent tissues,and the relationship between ITGα5 expression and clinicopathological parameters was analyzed.According to the different transfected substances in HEC-1B cells,they were divided into Control group(without any treatment),NC group(transfected si-NC)and ITGα5 group(transfected siRNA-ITGα5).Western blot was used to detect the protein expression levels of ITGα5 in each group before and after transfection.The expression level of ITGα5-mRNA was detected by real-time fluorescence quantitative polymerase chain reaction(qRT-PCR).MTT assay was used to detect cell proliferation,flow cytometry was used to detect cell cycle,Transwell chamber assay was used to detect cell invasion,and scratch assay was used to detect cell migration.Results The positive rate of ITGα5 in endometrial carcinoma tissues(58.89%)was higher than that in adjacent tissues(13.33%)(P=0.000).The positive rate of ITGα5 was correlated with pathological type,pathological stage,degree of differentiation and lymph node metastasis(P=0.000).The expression levels of ITGα5-mRNA and ITGα5 protein of HEC-1B cells were decreased after ITGα5 silencing(P<0.05),and the proliferation,migration and invasion abilities of HEC-1B cells were significantly decreased(P<0.01).Conclusions ITGα5 is highly expressed in endometrial carcinoma.Silencing ITGα5 can inhibit the proliferation,migration and invasion of HEC-1B cells,suggesting that ITGα5 plays an important role in the occurrence and development of endometrial carcinoma.
