宫颈癌患者外周血LGR5水平与肿瘤增殖、侵袭基因表达的相关性 被引量:1

摘要 目的探究宫颈癌患者外周血富含亮氨酸重复序列的G蛋白偶联受体5(LGR5)水平与肿瘤增殖、侵袭基因表达的相关性。方法选取2019年4月至2020年4月宜宾市第一人民医院诊治的148例宫颈癌患者作为研究对象,分为早期组(Ⅰ期,n=35)、中期组(Ⅱ~Ⅲ期,n=75)和晚期组(Ⅳ期,n=38)。另选择同期进行宫颈检查的50例宫颈息肉患者作为对照组。比较四组LGR5蛋白及侵袭基因、增殖基因水平,采用Pearson相关性分析患者外周血LGR5水平与肿瘤增殖、侵袭基因表达的相关性。结果早期组、中期组、晚期组的LGR5水平均显著高于对照组,且晚期组LGR5水平显著高于早期组、中期组,中期组LGR5水平显著高于早期组,差异具有统计学意义(P<0.05);早期组、中期组、晚期组的肿瘤组织增殖基因CD59、SRPX2、TBX2、NDRG3 mRNA表达量以及侵袭基因hnRNPH、caspase-8、PRPS2、Rab11 mRNA表达量均显著高于癌旁正常组织,差异具有统计学意义(P<0.05);晚期组肿瘤组织增殖基因、侵袭基因表达量均显著高于早期组、中期组,中期组肿瘤组织增殖基因、侵袭基因表达量显著高于早期组,差异具有统计学意义(P<0.05);经Pearson相关性分析发现,外周血LGR5水平与宫颈癌患者肿瘤组织增殖基因CD59、SRPX2、TBX2、NDRG3 mRNA水平以及侵袭基因hnRNPH、caspase-8、PRPS2、Rab11 mRNA均呈正相关(P<0.05)。结论外周血中LGR5表达越高,宫颈癌分期越高,且肿瘤细胞增殖、侵袭等恶性行为越活跃,检测LGR5有助于宫颈癌患者的诊断及预后评估。 Objective To explore the correlation between the level of leucine rich G protein coupled receptor 5(LGR5)in peripheral blood of patients with cervical cancer and tumor proliferation and invasion genes expression.Methods 148 patients with cervical cancer treated in First People′s Hospital of Yibin City from April 2019 to April 2020 were selected and divided into early group(stageⅠ,n=35),intermediate group(stageⅡtoⅢ,n=75)and advanced group(stageⅣ,n=38).Another 50 patients with cervical polyps who underwent cervical examination at the same period were selected as the control group.The levels of LGR5 protein,invasive genes and proliferative genes in the four groups were compared.Pearson correlation was used to analyze the correlation between the levels of LGR5 in the patients′peripheral blood and the expression of tumor proliferation and invasion genes.Results The level of LGR5 in early group,intermediate group and advanced group was significantly higher than that in control group,and the level of LGR5 in advanced group was significantly higher than that in early group and intermediate group,and the level of LGR5 in intermediate group was significantly higher than that in early group,the difference was statistically significant(P<0.05).The mRNA expression levels of proliferation genes CD59,SRPX2,TBX2 and NDRG3 and invasion genes hnRNPH,caspase-8,PRPS2 and Rab11 in tumor tissues of early,intermediate and advanced groups were significantly higher than those in adjacent normal tissues,the difference was statistically significant(P<0.05).The expression levels of proliferative genes and invasive genes in tumor tissues in advanced group were significantly higher than those in early and intermediate groups,and the expression levels of proliferative genes and invasive genes in intermediate group were significantly higher than those in early group,with statistical significance(P<0.05).Pearson correlation analysis showed that LGR5 levels in peripheral blood were positively correlated with the mRNA levels of proliferation genes CD59,SRPX2,TBX2 and NDRG3,as well as the mRNA levels of invasion genes hnRNPH,caspase-8,PRPS2 and Rab11 in cervical cancer patients(P<0.05).Conclusions The higher the expression of LGR5 in the peripheral blood,the higher the stage of cervical cancer,and the more malignant behaviors such as tumor cell proliferation and invasion.The detection of LGR5 is helpful for the diagnosis and prognosis evaluation of cervical cancer patients.
