环状RNA泛素关联蛋白2在子宫内膜癌中的表达及其与患者预后的关系

The expression of circ RNA UBAP2 in endometrial carcinoma and its relationships with patients′prognosis

目的探究环状RNA(circ RNA)泛素关联蛋白2(UBAP2)在子宫内膜癌中的表达及其与患者病理特征和预后的关系。方法选取2017年3月至2020年3月海南省儋州市中医医院收治的82例子宫内膜癌患者的手术切除标本为子宫内膜癌组,选取同期因子宫肌瘤或子宫脱垂等行手术治疗的82例患者的正常子宫内膜标本为对照组。采用实时荧光定量聚合酶链反应(qRT-PCR)法检测circ RNA UBAP2表达水平;随访子宫内膜癌患者3年生存情况;COX回归分析影响子宫内膜癌患者预后的危险因素。结果与对照组相比,子宫内膜癌组circ RNA UBAP2表达水平显著下降(P<0.05);circ RNA UBAP2水平与患者组织学分级、病理分期、有无淋巴结转移、子宫肌层浸润深度有关(P<0.05);生存分析发现,circ RNA UBAP2低表达的子宫内膜癌患者中位生存时间为13.32个月,显著低于circ RNA UBAP2高表达的患者(22.43个月)(P<0.05);COX回归分析发现,子宫肌层浸润深度>1/2、有淋巴结转移、circ RNA UBAP2低表达是子宫内膜癌患者预后不良的独立危险因素(P<0.05)。结论circ RNA UBAP2在子宫内膜癌组织中呈低表达,与患者组织学分级、病理分期、有无淋巴结转移、子宫肌层浸润深度及3年生存情况有关,可能成为子宫内膜癌患者预后的临床监测指标。

Objective To investigate the expression of circular RNA(circ RNA)ubiquitin associated protein 2(UBAP2)in endometrial carcinoma(EC)and its relationships with clinicopathological features and prognosis.Methods The specimens of 82 patients underwent endometrial carcinoma surgery in Danzhou Hospital of Traditional Chinese Medicine in Hainan province from March 2017 to March 2020 were selected as endometrial carcinoma group,and 82 normal endometrial specimens from patients treated by surgery for uterine fibroids or uterine prolapse during the same period were selected as the control group.Circ RNA UBAP2 expression was detected by real-time fluorescence quantitative polymerase chain reaction(qRT-PCR).Patients with endometrial carcinoma were followed up for 3 years.COX regression was used to analyze prognostic risk factors in endometrial carcinoma patients.Results Compared with the control group,the expression level of circ RNA UBAP2 in endometrial carcinoma group was significantly decreased(P<0.05).The level of circ RNA UBAP2 was correlated with histological grade,pathological stage,lymph node metastasis and myometrium invasion depth(P<0.05).Survival analysis showed that the median survival time of endometrial carcinoma patients with low expression of circ RNA UBAP2 was 13.32 months,which was significantly lower than that of patients with high expression of circ RNA UBAP2(22.43 months)(P<0.05).COX regression analysis showed that the depth of myometrium invasion>1/2,lymph node metastasis and low expression of circ RNA UBAP2 were independent risk factors for poor prognosis of endometrial carcinoma patients(P<0.05).Conclusions The low expression of circ RNA UBAP2 in endometrial carcinoma tissues is related to the histological grade,pathological stage,lymph node metastasis,myometrial invasion depth and 3-year survival of patients,and may be a clinical monitoring index for the prognosis of patients with endometrial carcinoma.
