摘要
目的运用网络药理学的研究方法预测二妙散治疗腰椎管狭窄症(lumbar spinal stenosis,LSS)早期腰背部疼痛相关的有效成分、作用靶点及分子机制。方法首先以二妙散中"黄柏""苍术"作为检索对象,筛选出其有效成分及作用靶点;然后以"腰椎管狭窄症"为关键词,寻找疾病的相关靶点并借助韦恩图,得到药物-有效成分-疾病靶点的合集;随之借助string在线平台构建蛋白互作网络;最后应用R语言软件,完成富集分析。结果经筛选后,得到二妙散有效成分46个、作用靶点189个,而与腰椎管狭窄症相关的靶点共1829个;根据韦恩图得到二妙散作用于腰椎管狭窄症的靶点共93个;GO功能富集分析得到生物过程2051条,分子功能173项,细胞组分48项。而KEGG通路富集分析得到153条信号通路。结论二妙散主要通过槲皮素、汉黄芩素、豆甾醇等作用于JUN、MYC、FOS、TP53、MAPK14、ESR1等靶基因,经过对氧化应激的反应、对营养水平的反应、对脂多糖的反应及对类固醇激素的反应等生物过程,同时根据图表及查阅相关文献,认为KEGG通路分析主要依靠AGE-RAGE信号通路、IL-17信号通路、流体剪切应力与动脉粥样硬化、松弛素信号通路、TNF信号通路等发挥抗炎、抗氧化、调节免疫等作用。
Objective To predict the effective components,targets and molecular mechanisms of Ermiao San(二妙散)in the treatment of lumbar spine stenosis(LSS)in early stage by using network pharmacology.Methods First,“Huangbo”and“Cangzhu”in Ermiao San were selected as the search objects,and their effective components and action targets were screened out.Then,with“lumbar spinal stenosis”as the key word,the relevant targets of the disease were searched and the collection of drug-active ingredients-disease targets was obtained with the help of Venn diagram.the protein interaction network was constructed by the STRING online platform.Finally,R language software was used to complete the enrichment analysis.Results After screening,46 active components and 189 targets of Ermiao San were obtained,and 1829 targets related to lumbar spinal stenosis were obtained.According to the Venn diagram,a total of 93 targets of Ermiao San for lumbar spinal stenosis were obtained.Go functional enrichment analysis obtained 2051 biological processes,173 molecular functions and 48 cellular components.The KEGG pathway enrichment analysis obtained 153 signaling pathways.Conclusion Ermiao San mainly acts on target genes such as JUN,MYC,FOS,TP53,MAPK14 and ESR1 through quercetin,wogonin,beta-sitosterolβand stigmasterol.Through biological processes such as responses to oxidative stress,nutrient levels,lipopolysaccharides,and steroid hormones,and based on charts and literature review.It is suggested that KEGG pathway analysis mainly relies on AGE-RAGE signaling pathway,IL-17 signaling pathway,fluid shear stress and atherosclerosis,relaxin signaling pathway,TNF signaling pathway and other pathways to play anti-inflammatory,anti-oxidation,immune regulation and other functions.
