摘要
本研究旨在建立一种寨卡病毒的RPA-LFD检测体系,达到简捷、灵敏、特异的检测目的,更好地应用于口岸卫生检疫及基层现场检测。通过引物设计筛选和反应条件的优化,建立寨卡病毒RPA-LFD检测技术,并对其灵敏性、特异性进行评估。通过检测41例临床样本,对检测体系的可行性及效果进行评估。检测结果具有特异性,与常见病原无交叉反应;与荧光定量PCR(qPCR)技术、PCR技术进行比较,具有很好的灵敏度,且可通过肉眼判断检测结果。结果显示,该方法可弥补PCR技术无法进行现场筛查的缺陷,非常适合口岸卫生检疫及基层现场检测,具有广阔的应用前景。
The aim of this study was to establish a simple,sensitive and specific RPA-LFD detection system for Zika virus,which could be better used in port health quarantine and frontline detection.By screening primer design and optimizing reaction conditions,the RPA-LFD detection technique for Zika virus was established,and its sensitivity and specificity were evaluated.In addition,41 clinical samples were tested to evaluate the feasibility and effectiveness of the detection system.Compared with fluorescence quantitative PCR(qPCR)and PCR,the results of RPA-LFD showed good sensitivity in the detection of Zika virus and no cross-reaction with com mon pathogens,and the results could be judged by the human eye.The results showed that it was very suitable for port health quarantine and frontline detection,with broad prospect for application.
