白藜芦醇在对乙酰氨基酚诱导巨噬细胞损伤中的修复作用 被引量:1

摘要 目的评价对乙酰氨基酚对巨噬细胞的损伤效果,研究白藜芦醇对损伤细胞干预和修复机制。方法利用对乙酰氨基酚构建RAW264.7细胞损伤模型,通过MTT以及Hoechst 33258染色分析细胞增殖和凋亡情况,检测给药后细胞中过氧化氢酶(CAT)、超氧化物歧化酶(SOD)、诱导型一氧化氮合酶(iNOS)活性变化及谷胱甘肽(GSH)与一氧化氮(NO)水平。结果对乙酰氨基酚可以抑制巨噬细胞增殖,促进细胞凋亡,诱发巨噬细胞发生氧化应激反应和炎症反应,促进细胞产生炎症因子IL-1β、IL-6及NO水平(P<0.01),提高细胞中iNOS酶活性(P<0.01)。白藜芦醇能修复损伤细胞中抗氧化体系,上调CAT、SOD及GSH活性(P<0.05,P<0.01),抑制iNOS和NO表达(P<0.05,P<0.01),提高细胞的生存率(P<0.05,P<0.01)。结论对乙酰氨基酚对巨噬细胞具有细胞毒性,白藜芦醇可修复对乙酰氨基酚对巨噬细胞造成的细胞损伤,其修复作用可能与调控细胞中抗氧化酶活性和炎症因子水平有关。 AIM To assess the damaging effects of acetaminophen(APAP) on macrophage and to investigate the mechanism of resveratrol in repair of cell injury. METHODS After being induced into injury models by APAP, the RAW264.7 cells were subjected to the analyses of their proliferation and apoptosis by MTT and Hoechst 33258 staining;and the determination of their activity changes of CAT,SOD,iNOS and the levels of GSH and NO as well. RESULTS The macrophages intervened with APAP demonstrated inhibited cell proliferation, promoted cell apoptosis, promoted oxidative stress and inflammatory responses, and levels of inflammatory factor IL-1β, IL-6 and NO(P<0.01),and increased iNOS enzyme activity in cells(P<0.01). Resveratrol repaired the antioxidant system in injured cells, up-regulated CAT,SOD,and GSH activities(P<0.05,P<0.01),down-regulated iNOS and NO levels(P<0.05,P<0.01),and improved cell survival(P<0.05,P<0.01).CONCLUSION Resveratrol can repair macrophages injury induced by cytotoxic APAP, with its efficacy in the regulation of the activities of antioxidant enzymes and the levels of inflammatory factors in RAW264.7 cells.
