摘要
用酶联免疫法检测柴胡口服液作用下5 d和10 d热应激肉鸡血清中炎性因子的表达,用蛋白免疫印迹法检测肉鸡肺脏中炎症相关因子(NF-κB p65、NLRP3、IL-1β、IL-6和TNF-α)蛋白表达,并观察肺脏病理切片。发现慢性热应激5 d与10 d显著提高了肉鸡血清中促炎因子IL-1β和IL-6、TNF-α的含量,肉鸡出现肺泡结构损伤、充血等病理变化,而柴胡口服液能明显降低第5天血清中促炎因子IL-1β的含量,降低第10天血清中IL-6和TNF-α的水平,改善肺组织病理损伤,并能显著下调肺组织炎性因子NF-κB p65、NLRP3、IL-1β、IL-6和TNF-α的蛋白表达水平。说明慢性热应激可诱导肉鸡肺脏炎性损伤,柴胡口服液可通过下调NF-κB-NLRP3信号通路抑制炎性因子蛋白表达,从而缓解热应激诱导的肺损伤。
ELISA was used to detect the serum inflammatory factor levels in Radix bupleuri oral liquid for 5 or 10 days treated chickens with chronic heat stress.Moreover,Western blot was used to detect the protein expressions of pulmalary inflammatory related factors(NF-κB p65,NLRP3,IL-1β,IL-6 and TNF-α),and the histopathological lesions of lung tissue was observed.Results showed that chronic heat stress for 5 d and 10 d significantly improved the chicken serum levels of pro-inflammatory factors IL-1β,IL-6 and TNF-αrespectively.It also can induce chicken alveolar structure damage,congestion and other pathological lesions.Radix bupleuri oral liquid for 5 d can significantly reduce serum IL-6 level,for 10 d can decrease pro-inflammatory factor levels of IL-6 and TNF-α,and alleviate the pulmalary pathological damages.Protein expressions of NF-κB p65,NLRP3,IL-1β,IL-6 and TNF-αin lung were obviously inhibited by Radix bupleuri oral liquid.These results suggested that chronic heat stress can induce pulmalary inflammatory injury in broilers,and Radix bupleuri oral liquid can alleviate lung injury induced by heat stress through down-regulating NF-κB-NLRP3 signaling pathway to inhibit the inflammatory factor expressions.
