基于HepG2细胞及斑马鱼模型的瓜蒌薤白类方抗高脂血症药理作用比较研究 被引量:5

Comparative study on the anti-hyperlipidemia effects of Gualou Xiebai Categorized Formula based on HepG2 cells and zebrafish model

摘要 目的:研究瓜蒌薤白半夏汤(GXB)与瓜蒌薤白汤(GX)的抗高脂血症药理作用,并比较二方作用的强弱。方法:通过油酸诱导HepG2细胞脂质沉积模型,测定细胞内三酰甘油(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-c)、高密度脂蛋白胆固醇(HDL-c)、脂质的含量;通过喂食高胆固醇饲料建立Fli1:eGFP斑马鱼高脂血症模型,测定斑马鱼TG、TC、LDL-c、HDL-c的含量,观察斑马鱼血管中胆固醇积累的情况。结果:与模型组相比,GXB、GX均可显著降低HepG2细胞内TG、TC、LDL-c、脂质的含量,升高HDL-c的含量(P<0.05);GXB、GX均可显著降低斑马鱼TG、TC、LDL-c的含量,升高HDL-c的含量,抑制血管中胆固醇的积累(P<0.05);并且GXB的效应强度优于相同剂量水平的GX。结论:GXB、GX可调节血脂水平,抑制脂质积累,具有较好的抗高脂血症药理作用,增加半夏的GXB,其抗高脂血症药理作用优于GX,为临床应用于防治高脂血症和开发高效、针对性强的抗高脂血症新药提供理论依据。 OBJECTIVE To compare the anti-hyperlipidemia effects between Gualou Xiebai Banxia Decoction(GXB) and Gualou Xiebai Decoction(GX), and compare their effect intensity.METHODS The lipid accumulation HepG2 cell model was induced by oleic acid, the contents of triglyceride(TG), total cholesterol(TC), low density lipoprotein cholesterol(LDL-c), high density lipoprotein cholesterol(HDL-c) and lipid were determined in HepG2 cells. The hyperlipidemia zebrafish model was established by feeding with high cholesterol diet, the contents of TG, TC, LDL-c and HDL-c were determined in zebrafish, and the accumulation of cholesterol in zebrafish blood vessels was observed.RESULTS Compared with the model group, GXB and GX could significantly reduce the contents of TG, TC, LDL-c and lipid, and increase the content of HDL-c in HepG2 cells(P<0.05). GXB and GX could significantly reduce the contents of TG, TC and LDL-c, increase the content of HDL-c in zebrafish, and inhibit the accumulation of cholesterol in zebrafish blood vessels(P<0.05). In addition, the effect intensity of GXB was better than that of GX at the same dose level.CONCLUSION GXB and GX have good anti-hyperlipidemia pharmacological effects through regulating lipid level and inhibiting the accumulation of lipid. The anti-hyperlipidemia activity of GXB is increased by Banxia, which is better than that of GX. The study provides theoretical basis for the clinical application in the prevention and treatment of hyperlipidemia and the development of highly effective and targeted new anti-hyperlipidemia drugs.
