泛素结合酶E2在肝细胞癌中的机制研究进展

摘要 泛素化在蛋白降解过程中发挥重要作用,参与基因转录和翻译、信号转导和细胞周期调控等生物学过程。泛素化由泛素激活酶E1-泛素结合酶E2-泛素连接酶E3级联依次介导。泛素结合酶E2可促进泛素与底物的结合,调节各种蛋白的稳定性以及蛋白间的相互作用。在肝细胞癌中,泛素结合酶E2参与DNA修复、细胞凋亡及细胞周期调节等,并可激活促癌信号通路,促进肝细胞癌进展。本文就泛素化、泛素结合酶E2的类型、与肝细胞癌有关的泛素结合酶E2的研究进展作一综述。 Ubiquitination plays an important role in protein degradation and is involved in biological processes such as gene transcription and translation, signal transduction, and cell cycle. The process of ubiquitination is mediated by the E1(activating)-E2(conjugating)-E3(ligating) enzyme cascade. E2 enzymes can promote the conjugation of ubiquitin to substrates, regulate the stability of various proteins and protein-protein interactions. E2 enzymes are involved in various pro-oncogenic processes of hepatocellular carcinoma, including DNA repair, regulation of apoptosis, regulation of cell cycle progression and activation of oncogenic signaling pathways. This paper reviews the research progress of ubiquitination, E2 enzyme types, and hepatocellular carcinoma related E2 enzymes.
