硫酸铵替代硝酸铵对天胡荽组织培养不定芽的影响 被引量:1

摘要 以天胡荽不定芽为试材,采用组织培养方法,研究了硫酸铵对天胡荽不定芽增殖的影响,为MS培养基NH4NO_(3)替代提供依据。结果表明:(NH4)2 SO_(4)1360 mg·L^(-1)+KNO_(3)2080 mg·L^(-1)作为MS培养基中NH4NO_(3)的替代物有利于天胡荽不定芽增殖。改良MS((NH_(4))_(2)SO_(4)1360 mg·L^(-1)+KNO_(3)2080 mg·L^(-1))+NAA 0.1 mg·L^(-1)+6-BA 2 mg·L^(-1)培养的天胡荽不定芽生长质量倍增率(1.87)、株高增量(1.30 cm)、增殖有效芽倍增率(2.16)、生根有效芽倍增率(1.70)均显著高于对照(MS+NAA 0.1 mg·L^(-1)+6-BA 2 mg·L^(-1));增殖培养20 d后,可溶性蛋白质含量(9.25 mg·g^(-1))和可溶性糖含量(13.19 mg·g^(-1))与对照无显著性差异;硝酸还原酶活性(NaNO_(3)23.11μg·g^(-1)·h^(-1))极显著地高于其他处理。(NH_(4))_(2)SO_(4)可以替代MS培养基中的NH4NO_(3),最适浓度为(NH_(4))_(2)SO_(4)1360 mg·L^(-1)+KNO_(3)2080 mg·L^(-1)。 In the study,adventitious buds of coriander were used for tissue culture to explore the effects of ammonium sulfate on proliferation of adventitious buds in order to provide the basis for the substitution of NH_(4)NO_(3)in MS medium.The results indicated that(NH_(4))_(2) SO_(4)1360 mg·L^(-1)+KNO_(3)2080 mg·L^(-1)as substitutes of NH_(4)NO_(3)in MS medium were beneficial to adventitious bud proliferation of coriander.It was shown in the following aspects.The rate of growth quality(1.87),the plant height increment(1.30 cm),the rate of available proliferation buds(2.16)and the rate of available rooting buds(1.70)of the adventitious buds of coriander culture with improved MS((NH_(4))_(2) SO_(4)1360 mg·L^(-1)+KNO_(3)2080 mg·L^(-1))+NAA 0.1 mg·L^(-1)+6-BA 2 mg·L^(-1)were significantly higher than those in control MS medium(MS+NAA 0.1 mg·L^(-1)+6-BA 2 mg·L^(-1)).After 20 days of proliferation culture,there was no significant differences in content of soluble protein(9.25 mg·g^(-1))and soluble sugar(13.19 mg·g^(-1))between(NH_(4))_(2)SO_(4)1360 mg·L^(-1)+KNO_(3)2080 mg·L^(-1)as substitute of NH_(4)NO_(3)and MS medium.The activity of nitrate reductase(NaNO_(3)23.11μg·g^(-1)·h^(-1))was very significantly higher than that under other treatments.The optimal concentrations of(NH_(4))_(2)SO_(4)as substitute of NH_(4)NO_(3)in MS medium were(NH_(4))_(2)SO_(4)1360 mg·L^(-1)+KNO_(3)2080 mg·L^(-1).
