摘要
目的:明确血清长链非编码RNA(LncRNA)miR155 Host Gene(LncRNA miR155HG)在肺腺癌(LUAD)患者血清中的表达,探讨其表达水平与临床特征的相关性,评价血清中LncRNA miR155HG作为LUAD生物标志物的可能性及优越性。方法:选择LUAD患者38例,留取患者空腹状态下血清4 ml。同时期留取健康对照组血清25例。采用实时荧光定量PCR(RT-qPCR)方法检测LncRNA miR155HG在血清标本中的表达水平。实验中以GAPDH作为内参照。结果:lncRNA miR155HG在LUAD患者血清中的表达水平较健康对照组高(P<0.0001)。LncRNA miR155HG表达水平与LUAD临床特征如年龄、性别、肿瘤直径、淋巴结转移情况、远端转移情况等无相关性(均P>0.05)。LncRNA miR155HG作为LUAD诊断生物标志物时,ROC曲线下面积、灵敏度及特异度分别为0.8105,91.56%、70.55%。生存分析结果显示,高表达患者预后优于低表达患者,但差异无统计学意义(均P>0.05)。结论:血清LncRNA miR155HG具有作为LUAD诊断生物标志物的可能,其具有无创、便捷、稳定的优点,本研究为LUAD的早期诊断、早期治疗提供新的线索。
Objective:To determine the expression of long non-coding RNA(lncRNA)miR155 Host Gene(lncRNA miR155HG)in serum of patients with lung adenocarcinoma(LUAD),and to explore the correlation between its expression level and clinical characteristics,and to evaluate the possibility and superiority of serum lncRNA miR155HG as a biomarker of LUAD.Methods:38 LUAD patients were selected and serum was collected in fasting condition.At the same time,serum of 25 healthy controls were collected.Quantitative real-time PCR(RT-qPCR)was used to detect the expression of lncRNA miR155HG in serum.GAPDH was used as internal reference in the experiment.Results:The expression level of lncRNA miR155HG in serum of LUAD patients was higher than that of healthy controls(P<0.0001).There was no correlation between the expression of lncRNA miR155HG and the clinical characteristics of LUAD patients,such as age,sex,tumor diameter,lymph node metastasis and distal metastasis(all P>0.05).When lncRNA miR155HG was used as biomarker of LUAD,the area under ROC curve was 0.8105,the sensitivity was 91.56%,and the specificity was 70.55%.Survival analysis showed that the prognosis of patients with high expression was better than that of patients with low expression,but the difference was not statistically significant(all P>0.05).Conclusion:LncRNA miR155HG in serum is a potential biomarker for the diagnosis of LUAD,which has the advantages of noninvasive,convenient and stable.This study provides new clues for the early diagnosis and treatment of LUAD.
