摘要
为探讨斑玉蕈(Hypsizygus marmoreus)交配型位点结构及其多态性,通过制备双核菌株H3、X3和B3的原生质体及再生获得其单核菌株,分别命名为H3-1、X3-1、B3-1,其他按顺序编号;H3-1、X3-1、B3-1分别与同一菌株的其他单核菌株两两配对,各选取1对亲和单核菌株,两两配对测定交配型,采用生物信息学分析比较基因组重测序后不同单核菌株的A和B交配型位点结构及其多态性。结果表明:双核菌株H3、X3、B3分别分离得到2、13、5对亲和单核菌株,各选取的1对亲和单核菌株两两配对,确定6个单核菌株H3-1、H3-8、X3-1、X3-13、B3-1、B3-2的交配型分别为A1B1、A2B2、A1B3、A3B1、A4B1、A5B4;6个单核菌株的A交配型位点两翼紧邻MIP和B-fg保守基因,且HD2.2结构域共线性良好,单核菌株H3-8和B3-1的A交配型位点结构完全相同,6个单核菌株共包括5种A交配型位点结构,表明A交配型位点具有丰富的多态性;单核菌株X3-1包含5个信息素受体,单核菌株X3-13、H3-1、B3-2均包含7个信息素受体,单核菌株H3-8、B3-1均包含6个信息素受体,6个单核菌株B交配型位点共包括4种,单核菌株H3-1、X3-13、B3-1、H3-8、B3-2均包含1个信息素前体,单核菌株X3-1包含2个信息素前体,B交配型位点结构上下游基因的共线性较差,表明B交配型位点具有丰富的多态性。
The genetic structure and polymorphism of mating-type loci in Hypsizygus marmoreus were studied through sequencing six monokaryotic strains segregated from three dikaryotic H. marmoreus strains, H3, X3 and B3, respectively. The six monokaryotic strains were H3-1, H3-8, X3-1, X3-13, B3-1 and B3-2. Each pair of monokaryotic strains derived from the same dikaryotic strain were compatible and contained different mating types. The results showed that there were five kinds of A mating type locus structures and four kinds of B mating type locus structures in the six monokaryotic strains, respectively. The mating types of H3-1, H3-8, X3-1, X3-13, B3-1 and B3-2 were A1B1, A2B2, A1B3, A3B1, A4B1 and A5B4, respectively. H3-8 and X3-1 had the same A mating type locus structure. All the A mating-type loci were flanked by conserved MIP and B-fg at the two ends respectively, and had a tight collinearity in HD2.2 domain. For B mating type loci, there were five pheromone receptors in X3-1, six pheromone receptors in H3-8 and B3-1, and seven pheromone receptors in X3-13, H3-1 and B3-2. There was one pheromone precursor in H3-1, X3-13, B3-1, H3-8 and B3-2. X3-1 contained two pheromone precursors. The collinearity between genes upstream and downstream of B mating type loci was poor, suggesting a rich genetic diversity.
