摘要
为探讨广东虫草(Cordyceps guangdongensis)子实体乙醇提取物对高脂饮食(high-fat diet,HFD)小鼠体质量和肠道菌群的调节作用,采用高脂饲养小鼠构建肥胖模型,灌胃广东虫草子实体乙醇提取物,比较肥胖小鼠的体质量和体内脂肪质量的变化;测定血清中甘油三酯(triglyceride,TG)、总胆固醇(total cholesterol,TC)和低密度脂蛋白胆固醇(low density lipoprotein cholesterin,LDL-C)等的含量;分析粪便中菌群结构和短链脂肪酸含量的变化;观察肝脏中脂肪积累与病变情况。结果表明:与高脂组小鼠相比,灌胃广东虫草子实体乙醇提取物显著减轻高脂饮食诱导肥胖小鼠的体质量和脂肪质量,降低其血清中TG、TC和LDL-C的含量,减少肝脏质量并缓解其中脂滴积累和病变程度,显著降低肠道菌群中厚壁菌门(Firmicutes)和拟杆菌门(Bacteroidetes)丰度的比值,提高粪便中丁酸含量。
The effects of Cordyceps guangdongensis fruiting body ethanol extract(CE) on body weight and gut microbiota of mice fed with a high-fat diet(HFD) were studied. Mice were first fed with HFD to induce obesity and then gavaged with CE. Obesity-related indices, including body weight, fat mass, triglyceride(TG), total cholesterol(TC) and low-density lipoprotein cholesterol(LDL-C) were measured for both obese and CE treated mice. Gut microbiota and fecal short-chain fatty acid level were determined for both obese and CE treated mice. Fat accumulation and pathological changes in mouse liver were also observed. The results showed that CE significantly reduced the body weight and fat mass of HFD induced obese mice;decreased serum TG, TC and LDL-C;decreased liver weight and reduced lipid droplet accumulation, and obviously ameliorated hepatic lesions in obese mice. Compared with HFD fed mice, CE treated mice showed a significant decrease in the relative abundance of Firmicutes and Bacteroides and a significant increase in butyric acid content in feces.
