麻防犀角地黄汤对miR-215-5p的干预作用及对银屑病小鼠的影响

Effects of Mafang Xijiao Dihuang Decoction on the intervention of miR-215-5p and its influence on mouse model of psoriasis

目的:通过建立银屑病小鼠模型,观察麻防犀角地黄汤对银屑病皮损及miR-215-5p的影响,探讨麻防犀角地黄汤治疗银屑病的作用机制。方法:咪喹莫特(IMQ)乳膏局部涂抹诱导建立银屑病小鼠模型,麻防犀角地黄汤灌胃干预。观察小鼠体质量、PASI评分、左耳耳廓外缘的厚度、皮损组织的病理改变;qRT-PCR法检测皮损中miR-215-5p的表达水平。结果:与IMQ组比较,麻防犀角地黄汤组皮损明显改善,PASI评分下降(P<0.05),耳廓外缘厚度降低(P<0.05),体质量升高,组织病理显示表皮厚度和炎症细胞浸润程度均明显改善,皮损组织中miR-215-5p表达水平显著升高(P<0.05)。结论:麻防犀角地黄汤可减轻小鼠银屑病样皮损的严重程度、提高生存质量,其机制可能与上调miR-215-5p,进而靶向调控相关信号通路存在一定关系。

Objective: To observe the effects of Mafang Xijiao Dihuang Decoction on psoriasis skin lesions and miR-215-5p by establishing mouse model of psoriasis, and explore the mechanism of action of Mafang Xijiao Dihuang Decoction in the treatment of psoriasis. Methods: The mouse model of psoriasis was established by IMQ local application, and the treatment of Mafang Xijiao Dihuang Decoction was performed by gavage. The body weight, PASI score, the thickness of the outer margin of the left ear and the pathological changes of the skin lesions were observed. The expression level of miR-215-5p in skin lesions was detected by qRT-PCR. Results: Compared with the IMQ group, the skin lesions of the Mafang Xijiao Dihuang Decoction group were significantly improved, the PASI score decreased(P<0.05), the outer thickness of the auricle decreased(P<0.05), the body weight increased, and the histopathological findings showed that the epidermal thickness and the degree of inflammatory cell infiltration were significantly improved. The expression level of miR-215-5p in the skin tissues of the Mafang Xijiao Dihuang Decoction group was significantly higher than that of the IMQ group(P<0.05). Conclusion: Mafang Xijiao Dihuang Decoction can reduce the severity of psoriatic lesions and improve the quality of life in mice. The mechanism may be related to the up-regulation of miR-215-5p and the targeted regulation of related signaling pathways.
