摘要
[目的]对苋菜MDH基因进行克隆及生物信息学分析。[方法]以苋菜试管苗为材料,从实验室构建的苋菜转录组数据库中筛选出苋菜MDH基因的c DNA序列片段,采用RT-PCR技术对其ORF进行克隆,并进行生物信息学分析。[结果]MDH基因编码344个氨基酸,通过生物信息学分析表明,MDH可能为非分泌蛋白,包括33个磷酸化位点,二级结构以α螺旋为主,三级结构是由无规则卷曲结构、α螺旋和β折叠构成的,与藜麦有较近的进化关系,在黑暗条件下和叶片红色部位表达更显著开花期较其他生长时期表达更显著。[结论]为将来深入研究MDH基因功能奠定基础。
[Objective]To clone and bioinformatics of MDH gene in Amaranthus tricolor.[Method]Using amaranth test-tube seedlings as material,from the amaranth transcriptome database constructed in the laboratory,the cDNA sequence fragment of the MDH gene of amaranth was screened,and its ORF was cloned by RT-PCR and analyzed by bioinformatics.[Result]The results show that the MDH gene encodes 344 amino acids.Bioinformatics analysis shows that MDH may be a non-secreted protein,including 33 phosphorylation sites.The secondary structure is dominated by α helix,and the tertiary structure is composed of random coiled structures,Α-helix and β-sheet,it has a close evolutionary relationship with quinoa.[Conclusion]The expression is more significant in dark conditions and red parts of leaves,and more significant in flowering stage than in other growth stages.
