高癌家系气虚质鼻咽癌患者鼻咽组织标本RhoGDIβ的表达水平及其临床病理意义 被引量:1

Expression Level and Clinicopathological Significance of Nasopharyngeal Tissue RhoGDIβ in Patients with Nasopharyngeal Carcinoma with High Family Qi Deficiency

摘要 目的:研究RhoGDIβ蛋白在高癌家系气虚质鼻咽癌患者中的表达,并探讨其特异性体质病机。方法:于2019年11月—2020年11月期间,分别由我院就诊的散发性鼻咽癌和高癌家系鼻咽癌中选取气虚质患者各6例,取鼻咽病灶组织标本,并收集同期就诊的正常鼻咽组织标本6例,分别提取总RNA和蛋白质。应用蛋白免疫印迹(Western blotting)法分别检测各组组织标本RhoGDIβ蛋白表达水平,PCR法检测各组组织标本RhoGDIβRNA mRNA表达水平。结果:PCR检测显示散发性鼻咽癌组和高癌家系鼻咽癌组的RhoGDIβmRNA的表达水平明显高于正常组(P<0.05)。Western blot法检测RhoGDIβ蛋白结果显示,正常组与散发性鼻咽癌组和高癌家系鼻咽癌组比较,差异具有统计学意义(P<0.05)。进行组间两两比较,正常组与散发性鼻咽癌组、正常组与高癌家系鼻咽癌组比较差异均有统计学意义(P<0.05)。结论:无论是高癌家系鼻咽癌患者还是散发性鼻咽癌患者,气虚质患者鼻咽癌病灶组织中的RhoGDIβ表达均显示明显上调。该一指标有望成为早期诊断和筛查高癌家系气虚质鼻咽癌患者的客观标志物。 Objective:To study the expression of RhoGDIβ protein in patients with nasopharyngeal carcinoma(NPC) with hypercancerous qi deficiency and explore its specific constitution and pathogenesis.Methods:From November 2019 to November 2020,6 patients were selected from sporadic nasopharyngeal carcinoma and high-cancer nasopharyngeal carcinoma,take tissue samples from nasopharyngeal lesions,and collected 6 normal nasopharyngeal tissue samples,extraction of total RNA and proteins,respectively.The protein expression levels of tissue samples RhoGDIβ each group were detected by Western blotting method,the expression level of tissue samples RhoGDIβRNA mRNA each group was detected by PCR method.Results:PCR detection showed that the expression of RhoGDIβ mRNA in sporadic nasopharyngeal carcinoma group and high nasopharyngeal carcinoma group was significantly higher than that in normal group(P<0.05).The results of Western blotting detection of RhoGDIβ protein showed that the difference between normal group and sporadic nasopharyngeal carcinoma group and high nasopharyngeal carcinoma group was statistically significant(P<0.05).Compared with two groups,there was significant difference between normal group and sporadic nasopharyngeal carcinoma group,normal group and high cancer family nasopharyngeal carcinoma group(P<0.05).Conclusion:The expression of RhoGDIβ in the focal tissue of nasopharyngeal carcinoma was significantly up-regulated in patients with high or sporadic nasopharyngeal carcinoma.This index is expected to be an objective marker for early diagnosis and screening of patients with high cancer family qi deficiency nasopharyngeal carcinoma.
