摘要
探究白及提取物介导MAPK信号通路下调Caspase12对骨关节炎(OA)软骨细胞增殖和凋亡的影响。软骨细胞分为对照组、OA组、OA+白及组、OA+白及+SB203580组。通过10 ng/ml的IL-1β培养软骨细胞24 h以模拟体外OA,加入对应的含有50μg/ml的白及提取物预培养24 h,SB203580(5μmol/L)抑制p38 MAPK通路。检测各组细胞增殖、凋亡、细胞中Caspase-12、p38 MAPK蛋白表达水平。研究发现白及提取物可以通过抑制p38 MAPK通路抑制Caspase12的表达,从而缓解软骨损伤。
The purpose of this study was to investigate the effect of Bletilla striata extract on proliferation and apoptosis of chondrocytes in osteoarthritis(OA)by Caspase-12 inhibition through down-regulating the p38MAPK pathway.The studied chondrocytes were divided into the control group,OA+Bletilla striata group,OA+Bletilla striata+SB203580 group.Then incubated with 10 ng/mL IL-1βfor 24 h to simulate in vitro OA.The cells were pre-cultured for 24 h in the Bletilla striata extract containing 50μg/mL.SB203580(5μmol/L)was added to the culture medium to inhibit the p38 MAPK pathway.The chondrocyte proliferation and apoptosis,and protein expression levels of Caspase-12 and p38 MAPK in each group were examined.The study showed that Bletilla striata extract inhibited the expression of Caspase12 by down-regulating the p38 MAPK pathway,thereby alleviating cartilage damage.
