摘要
目的建立和评价泄泻食滞胃肠证小鼠模型,并用保和丸进行干预以验证模型并观察疗效。方法利用自食高蛋白高热量饲料联合灌胃植物油模拟饮食不节制备食滞胃肠泄泻小鼠模型,运用保和丸进行干预以验证模型,以宏观表征和微观指标进行综合评价。结果模型组小鼠出现便溏、秽臭、摄食量减少、体重增长缓慢等宏观表征以及粪便含水量升高(P<0.05),肠内纤维素酶、淀粉酶、乳糖酶活性降低(P<0.05),血清脂肪酶、胃泌素、胆囊收缩素水平下降(P<0.05)等微观理化指标改变,各宏观表征和微观指标与泄泻食滞胃肠证的诊断和辨证要点具有一定等效对应性;保和丸干预后模型小鼠粪便性状恢复正常,摄食量和体重稳定增加;与自然恢复组相比,中药干预组血清胃泌素显著升高(P<0.05)、脂肪酶和胆囊收缩素也有所上升(P>0.05);与正常组相比,自然恢复组多种消化酶活性显著上升,而中药干预组肠内消化酶活性下降至更接近正常组水平。结论采用自食高蛋白高热量饲料联合灌胃植物油模拟饮食不节病因可以成功建立泄泻食滞胃肠证小鼠模型,保和丸能上调血清胃泌素和胆囊收缩素水平,调节消化酶活性,改善泄泻食滞胃肠证小鼠消化机能。
Objective To establish and evaluate a mouse model of diarrhea with gastrointestinal food stagnation syndrome and the efficacy of Baohe Wan.Methods The model of diarrhea with gastrointestinal food stagnation syndrome was established by high-protein and high-calorie feed combined with gavage edible blended oil.Then,the model mice were treated with Baohe Wan to verify the model attributes.A comprehensive evaluation was made with macroscopic and microscopic indicators.Results The results showed that the model mice had macroscopic manifestations such as loose stool with a smell of rancid odor,decreased food consumption and slow bodyweight growth,as well as microscopic indicators change such as increased fecal water content(P<0.05),decreased activities of cellulase,amylase,and lactase(P<0.05),and decreased levels of serum lipase,gastrin and cholecystokinin(P<0.05).All macroscopic manifestations and microscopic indicators have equivalent correspondence with the diagnosis and syndrome differentiation essentials of diarrhea with gastrointestinal food stagnation syndrome.After treatment of Baohe Wan,the fecal traits of the model mice recovered to normal,and their food consumption and body weight were increased steadily.Compared with the natural recovery group,serum gastrin was significantly increased in the treatment group(P<0.05),and lipase and cholecystokinin were increased slightly(P>0.05).Compared with the normal group,the activities of various digestive enzymes in the natural recovery group were significantly increased,while the activities of digestive enzymes in the treatment group were decreased to a level closer to the normal group.Conclusion The mouse model of diarrhea with gastrointestinal food stagnation syndrome can be successfully established by using high-protein and high-calorie feed combined with gavage edible blended oil.Baohe Wan can up-regulate the levels of serum gastrin and cholecystokinin,regulate the activity of digestive enzymes and improve the digestive function of model mice.
