醒脑解郁胶囊对PSD模型大鼠海马CaM mRNA和CaMKII表达的影响 被引量:1

The effect of XingNao JieYu Capsule on PSD model rats of hippocampus CaMmRNA and CaMKII

摘要 目的观察醒脑解郁胶囊治疗卒中后抑郁(PSD)的可能作用机制。方法将120只SD大鼠随机分为空白组、空白中药组、空白阳性药组、模型组、中药早期干预组、阳性药早期干预组、中药治疗组、阳性药治疗组。各空白组不干预,其余5组采用大脑中动脉闭塞(MCAO)联合孤养法与腹腔注射利血平制备PSD大鼠模型。中药早期干预组与阳性药早期干预组MCAO术后,分别给予中药、阳性药连续灌胃35d;中药治疗组与阳性药治疗组PSD模型制备后,分别给予中药、阳性药灌胃21d。体重测量及糖水消耗试验、旷野试验等行为学评估大鼠抑郁状态,RT-PCR法检测大鼠海马CaMmRNA水平的影响,Western blot法分析大鼠海马组织中CaMKII表达的含量。结果与空白组相比,PSD造模成功后各组大鼠体重均明显下降(P<0.05,P<0.01),糖水消耗及旷野实验所测的运动得分显著降低(P<0.01);治疗后,与模型组相比,中药早期干预组、阳性药早期干预组、中药治疗组及阳性药治疗组上述指标均显著升高(P<0.05,P<0.01),中药早期干预组、阳性药早期干预组、中药治疗组、阳性药治疗组大鼠海马区CaMmRNA相对表达量均不同程度的增加(P<0.01,P<0.05)、CaMKⅡ表达均不同程度的增加(P<0.01)。结论醒脑解郁胶囊可增加PSD模型大鼠体重,改善行为学症状,上调PSD大鼠模型海马组织内CaMmRNA及CaMKII的表达。 Objective To observe the possible mechanism of Xingnao Jieyu capsule in treating post-stroke depression(PSD).Methods 120 SD rats were randomly divided into blank group,blank Chinese medicine group,blank positive medicine group,model group,Chinese medicine early intervention group,positive medicine early intervention group,Chinese medicine treatment group,positive medicine treatment group.Each blank group did not intervene,and the remaining 5 groups used middle cerebral artery occlusion(MCAO)combined with solitary culture and intraperitoneal injection of reserpine to prepare PSD rat model.Early intervention group of traditional Chinese medicine and early intervention group of positive drugs after MCAO,Chinese medicine and positive medicine were given 35 d of continuous gavage.After the preparation of post-stroke depression model,Chinese medicine and positive drugs were given 21 d.Behavioral assessment of depression in rats by weight measurement,sugar water consumption test,wilderness test.The effect of RT-PCR method on hippocampal CaMmRNA levels in rats,the content of CaMKII expression in rat hippocampal tissue was analyzed by Western blot method.Results Compared with the blank group,the body weight of each group decreased significantly(P<0.05,P<0.01)after successful PSD modeling,a significant decrease in sugar water consumption and motor scores measured in field experiments(P<0.01);After treatment,compared to the model group,Both the early intervention group of traditional Chinese medicine,the early intervention group of positive drugs,the treatment group of traditional Chinese medicine and the treatment group of positive drugs were significantly increased(P<0.05,P<0.01),the relative expression of CaMmRNA in hippocampus of rats was increased(P<0.01)in the early intervention group,the positive intervention group,the traditional Chinese medicine treatment group and the positive drug treatment group(P<0.05),CaMKⅡexpression increased in varying degrees(P<0.01).Conclusion Xingnao Jieyu capsule can increase body weight,improve behavioral symptoms and up-regulate the expression of CaMmRNA and CaMKII in hippocampus of PSD rats.
