不同干燥温度对潞党参药材中游离糖及多糖的影响 被引量:4

Effects of Different Drying Temperatures on the contents of Free Sugars and Polysaccharides in Codonopsi

摘要 目的建立基于亲水相互作用色谱-高效液相色谱法测定潞党参中游离果糖、蔗糖含量的方法,并考察不同温度干燥温度处理对潞党参药材中果糖、蔗糖以及多糖含量的影响。方法建立了亲水相互作用色谱-高效液相色谱-蒸发光散射检测器(HILIC-ELSD)的方法测定不同温度干燥的潞党参中果糖、蔗糖含量,用苯酚-硫酸法测定不同温度干燥的潞党参中多糖含量。结果果糖、蔗糖色谱峰能很好分离;果糖含量高低顺序:常温阴干≈40℃烘干>60℃烘干;蔗糖含量高低顺序:常温阴干>40℃烘干>60℃烘干;多糖含量高低顺序:60℃烘干>40℃烘干≈常温阴干。结论该HILIC-ELSD法操作简便,重复性、分离度好,可用于党参中果糖、蔗糖含量测定;不同温度干燥的潞党参中果糖、蔗糖及多糖含量存在差异,且显现出不同变化趋势,该研究结果可为潞党参药材质量评价提供科学参考。 Objective A quantitative analytical method of free saccharides including fructose and sucrose in Codonopsis pilosula by hydrophilic interaction liquid chromatography(HILIC)–HPLC was established.And,the effect on the contents of fructose,sucrose and polysaccharides in Codonopsis pilosula of different drying temperature treatments were investigated.Methods A HILIC-HPLC-evaporative light scattering detector(ELSD)method was established to determine the contents of fructose and sucrose in Codonopsis pilosula of different drying temperature treatments.The phenol-sulfuric acid method was used to determine the content of polysaccharide in Codonopsis pilosula of different drying temperature treatments.Results The chromatographic peaks of fructose and sucrose can be separated well;the order of fructose content in Codonopsis pilosula:shadow drying≈40℃ hot air drying>60℃ hot air drying;the order of sucrose content in Codonopsis pilosula:shadow drying>40℃ hot air drying>60℃ hot air drying;the order of polysaccharide content in Codonopsis pilosula:60℃ hot air drying>40℃ hot air drying≈shadow drying.Conclusion The HILIC-HPLC-ELSD method is easy to operate,repeatable,and has good resolution.It can be used to determine the contents of fructose and sucrose in Codonopsis pilosula;the contents of fructose,sucrose and polysaccharides in Codonopsis pilosula of different drying temperature treatments are different,and shows different trends.The results of this study can provide a scientific reference for the quality evaluation of Codonopsis pilosula.
