摘要
目的:探讨血浆中的尼曼-匹克病C2蛋白水平(NPC2)在脓毒症病理进展中的作用。方法:通过ELISA法检测和对比健康者、消化系统感染非脓毒症患者和消化系统感染脓毒症患者血浆中的NPC2浓度。采用盲肠结扎穿孔术建立小鼠脓毒症模型,检测重组NPC2蛋白对脓毒症小鼠死亡率的影响。结果:(1)女性健康者血浆中的NPC2水平显著低于男性健康者;(2)消化系统感染患者血浆中的NPC2浓度显著升高;(3)脓毒症患者血浆中的NPC2水平进一步升高,并随器官功能衰竭的加重逐步升高;(4)重组人或者小鼠NPC2蛋白对腹腔感染脓毒症小鼠的存活率没有显著影响。结论:研究结果说明,血浆中的NPC2水平对于不同器官感染所导致的脓毒症患者是反映器官功能障碍程度的有效指标,但并不在脓毒症病理进程中起关键作用。
Objective:To investigate the role of plasma Niemann Pick disease C2 protein level(NPC2)in the pathological progression of sepsis.Methods:The plasma concentrations of NPC2 in healthy subjects,patients with non sepsis and patients with sepsis were detected and compared by ELISA.The sepsis model of mice was established by cecal ligation and perforation,and the effect of recombinant NPC2 protein on the mortality of sepsis mice was detected.Results:①NPC2 concentration was significantly lower in healthy female than male;②The concentration of NPC2 in plasma of patients with digestive system infection increased significantly;③NPC2 concentration was further increased in sepsis patients,in sync with the degree of organ dysfunction;④intravenous administration of recombinant human and mouse NPC2 did not significantly change the mortality rate of septic mice.Conclusion:Combined with our previous findings,these results showed that plasma NPC2 is a useful indicator of organ dysfunction in sepsis patients caused by infections in different organ systems,but may not play a critical role in sepsis pathogenesis.
