水稻响应细菌性条斑病菌侵染的转录组分析 被引量:2

Transcriptome Analysis of Rice Responding to Infections of Xanthomonas oryzae pv.oryzicola Strains

摘要 由黄单胞杆菌的致病变种(Xoc)引起的水稻细菌性条斑病(BLS)是严重危害水稻产量的细菌性病害之一。我们利用转录组学分析方法研究水稻抗性近等基因系NIL-bls2应对高致病性野生型Xoc菌株gx01^(w)及无致病性突变菌株gx01^(w)侵染时的分子机制。结果表明:与非致病菌株gx01^(w)相比,致病菌株gx01^(w)侵染水稻叶片12 h后,在转录组水平共鉴定出415个差异表达基因(DEGs),分别为212个上调基因和203个下调基因。其中与植物抗性相关的基因涉及7个转录因子(MYB,ERF,WRKY,b HLH,MADS)、20个类受体蛋白激酶(WAK,CRK,NBS-LRR)、4个NBS-LRR受体蛋白、11个抗性相关蛋白(硫相关蛋白,CYP450蛋白)和3个糖基转移酶。GO功能富集分析发现303个DEGs,KEGG代谢途径富集分析发现79个DEGs参与苯丙氨酸生物合成、植物MAPK信号转导、植物激素信号转导、植物-病原互作等生物途径。本研究重点分析了不同致病性菌株侵染抗性近等基因系NIL-bls2的差异表达基因,阐述了NIL-bls2与不同致病性Xoc菌株间的分子互作机制,为探索Xoc菌株gx01^(w)的侵染机制和抗性基因bls2的抗病机制提供了新的数据资源和理论依据。 Bacterial leaf streak of rice(BLS)caused by Xanthomonas oryzae pv.oryzicola(Xoc)significantly reduce the yield of rice.Transcriptomic analysis was used to investigate the molecular mechanism of rice resistant near isogenic line NIL-bls2 responding the infection of highly pathogenic Xoc wild strain gx01^(w)and nonpathogenic mutant strain gx01^(w).The results showed that compared with the non-pathogenic mutant strain gx01^(w),415 differentially expressed genes(DEGs)were identified in the transcriptome data of rice leaves infected by pathogenic wild strain gx01^(w)at 12 hours of post inoculation(hpi),including 212 up-regulated genes and 203 downregulated genes.The genes related to plant resistance involved in 7 transcription factors(MYB,ERF,WRKY,b HLH,MADS),20 receptor-like kinases(WAK,CRK,NBS-LRR),4 NBS-LRR receptor proteins,11 resistance-related proteins(sulfur-related proteins,CYP450 proteins)and 3 glycosyltransferases.303 DEGs were identified in GO functional enrichment analysis,and 79 DEGs were identified in KEGG metabolic pathway enrichment analysis to participate in metabolic pathway such as phenylalanine biosynthesis,plant MAPK signal transduction,plant hormone signal transduction and plant-pathogen interaction,etc.The results showed the difference of DEGs in different pathogenic Xoc strains infecting rice resistant near isogenic line NIL-bls2,and expounded the mechanism of interactions between NIL-bls2 and different pathogenic Xoc strains,which provided new data resources and theoretical basis for studying the infection mechanism of Xoc strain gx01^(w)and the resistant mechanism of resistant gene bls2.
