利用SSR标记分析贵州中晚熟李资源遗传多样性 被引量:5

Genetic Diversity Analysis of Guizhou Middle-late Maturing Plums by SSR Markers

摘要 本研究利用SSR分子标记技术对贵州16份中晚熟李资源进行遗传多样性分析,以期为贵州中晚熟李种质资源遗传多样性评价、亲缘关系分析、种质保存与利用等提供理论依据。16对SSR引物共扩增得到119条谱带,其中多态性条带为118条,占总带数的99.16%,各引物的多态信息含量为0.483~0.876;16对引物在供试材料中等位基因数(Na)、有效等位基因数(Ne)、观察杂合度(Ho)、期望杂合度(He)以及Shannon’s多样性指数(I)的平均值分别为7.44、4.47、0.73、0.61和1.59,表明贵州中晚熟李资源具有较为丰富的遗传多样性;资源8460和3450A的相似系数最小,为0.113,5886和2553的相似系数最大,为0.941;在遗传相似系数0.328处,UPGMA聚类分析将供试的贵州中晚熟李资源划分为了四大类:7661单独为一类,1699、5206A为一类,3450A和3450B为一类,其余11份资源归为一类。结果表明,供试的16份贵州中晚熟李资源中,5886与2553遗传背景最为相似、亲缘关系最近,8460和3450A的亲缘关系最远。本研究基于SSR的分子标记技术,可用于揭示贵州中晚熟李资源的遗传多样性以及亲缘关系,为贵州中晚熟李种子资源的保存和利用提供一定的参考依据。 In order to provide th eoretical basis for genetic diversity evaluation,genetic relationship analysis,germplasm preservation and utilization,16 middle/late ripening plum resources that from different areas of Guizhou Province was analyzed by using SSR molecular marker technique.The results showed that,119 bands were amplified by 16 pairs of SSR primers,of which 118 were polymorphic bands,accounting for 99.16%.The polymorphism information content of SSR primers was 0.483~0.876.The mean values of allele(Na),effective allele(Ne),observed heterozygosity(Ho),expected heterozygosity(He)and Shannon’s diversity index(I)were7.44,4.47,0.73,0.61 and 1.59,respectively.Indicating that there was a relatively rich genetic diversity in Guizhou’s middle-late maturing plum resources.In addition,the similarity coefficient of resources between 8460and 3450A was the smallest(0.113),while the similarity coefficient between 5886 and 2553 was the largest(0.941).And at the coefficient of 0.328,UPGMA cluster analysis divided the 16 tested plum resources into four categories:7661 as a category alone,1699 and 5206A as a category,3450A and 3450B as a category,and the remaining 11resources as another category.Above all,among the 16 analyzed plum resources,the 5886 had the closest relationship with 2553,while 8460 and 3450A were furthest related.From the above,SSR molecular marker technology can be used to reveal the genetic diversity and relationship of middle/late maturing plum resources,and provide certain reference basis for the conservation and utilization of local middle/late maturing plum seed resources in Guizhou.
