摘要
目的研究紫胶中紫胶红色素的化学成分。方法采用开放硅胶柱色谱、ODS、HPLC等方法分离化合物,运用现代波谱技术确定化合物的结构。结果从紫胶甲醇提取物中分离纯化得到5个蒽醌类化合物,分别鉴定为红紫胶素(Erythrolaccin,1),1,3,4,6-四羟基-8-甲基蒽醌(1,3,4,6-tetrahydroxy-8-methyl-anthraquinone,2),去氧虫胶红素(deoxyerythrolaccin,3),2,3,5,6-四羟基-8-甲基蒽醌(2,3,5,6-tetrahydroxy-8-methyl-anthraquinone,4),胭脂红素(Aloesaponarin II,5)。结论化合物1~5为首次从紫胶中分离得到,并且1~4为首次报道核磁数据。同时,采用MTT法及PNGP法测定化合物1~5的细胞毒性和α-葡萄糖苷酶抑制活性。结果发现,5个化合物均无细胞毒性;化合物4,化合物5的α-葡萄糖苷酶抑制活性显著,IC50值分别为38.1μmol·L^(-1)和69.8μmol·L^(-1)。
Objective To study Lac dye chemical constituents from Lac.Methods The compounds were isolated by chromatography on silica gel column,ODS,HPLC and identified by physicalchemical properties and spectrascopic analysis.Results five compounds were obtained and identified as erythrolaccin(1),1,3,4,6-tetrahydroxy-8-methyl-anthraquinone(2),deoxyerythrolaccin(3),2,3,5,6-tetrahydroxy-8-methyl-anthraquinone(4),aloesaponarin II(5).Conclusion Compounds 1-5 were isolated from Lac for the first time,and compound 1-4 were reported NMR data for the first time.At the same time,the cytotoxicity andα-glucosidase inhibitory activity of compounds 1-5 were tested by MTT and PNGP method.The results showed that all of the five compounds had no cytotoxicity.The inhibitory activity of compound 4 and compound 5α-glucosidase was significant.The IC;were 38.1μmol·L^(-1)and 69.8μmol·L^(-1),respectively.
