白介素10启动子区单核苷酸多态性与口腔鳞状细胞癌相关性研究

The association between interleukin 10 gene promoter polymorphisms and oral squamous cell carcinoma risk

目的:研究白介素10(IL-10)基因rs1800871(-819T/C)、rs1800872(-592A/C)位点与中国汉族人群口腔鳞状细胞癌(OSCC)易感性及相关危险因素的关系。方法:共纳入125例OSCC患者(病例组)及110例年龄性别均匹配的对照人群(对照组),提取研究对象基因组DNA进行聚合酶链反应(PCR)后直接测序,分析并比较IL-10基因型在两组间的分布频率。结果:关联研究结果显示rs1800871位点TC基因型携带者较TT基因型、CC基因型携带者较TT+TC基因型,以及C等位基因型携带者OSCC发病风险显著增加。rs1800872位点AC基因型携带者较AA基因型、CC基因型携带者较AA+AC基因型,以及C等位基因型携带者OSCC发病风险显著增加。分层分析结果表明男性、老年人(年龄≥60岁)、饮酒及不吸烟人群表现出更为显著的统计学相关性。结论:rs1800871及rs1800872位点变异与中国汉族人群OSCC疾病发生存在相关性,在基因位点变异与吸烟饮酒的协同作用下OSCC发病风险可能显著升高。

Objective:To investigate the relationship between the rs1800871 and rs1800872 polymorphisms in interleukin-10(IL-10)gene and the susceptibility of oral squamous cell carcinoma(OSCC),and to analyze the related risk factor with the risk of OSCC.Methods:The expression of two polymorphic sites(rs1800871 and rs1800872)of IL-10 gene in 125 OSCC patients and 110 controls was detected by PCR.The distribution frequency of IL-10 genotype between the two groups was analyzed and compared.Results:The results of association study showed that the risk of OSCC was significantly increased in rs1800871 TC genotype carriers compared with TT genotype,CC genotype carriers compared with TT+TC genotype,and C allele carriers.In addition,the results of association study showed that the risk of OSCC was significantly increased in rs1800872 AC genotype carriers compared with TT genotype,CC genotype carriers compared with AA+AC genotype,and C allele carriers.Stratified analyses showed more significant statistical associations among men,older adults(age≥60 years),drinkers,and non-smokers.Conclusion:Rs1800871 and rs1800872 gene promoter polymorphisms are correlated with the occurrence of OSCC in Han Chinese population.Under the synergistic effect of gene locus variation and smoking and drinking,the risk of OSCC may significantly increase.
