摘要
目的:检测辛伐他汀体内给药对大鼠根尖周炎模型病灶中的作用及其对人骨髓间充质干细胞(HMSCs)中Cyr61表达的影响。方法:构建32只SD大鼠牙齿根尖周炎模型,分为对照组(8只),模型观察组(8只)和辛伐他汀用药组(16只),分期处死后,通过Micro-CT扫描和HE染色法观察病灶情况;用辛伐他汀处理HMSC细胞,检测碱性磷酸酶(ALP)活性及Wnt、β-catenin和Cyr61蛋白的表达。结果:大鼠根尖周炎模型建立成功;辛伐他汀用药明显减小大鼠根尖病灶体积;经辛伐他汀处理的HMSCs ALP、Wnt、β-catenin和Cyr61蛋白的表达均增高。结论:辛伐他汀可通过Wnt信号通路增加Cyr61的表达并促进成骨。
Objective:To study the effects of simvastatin on periapical periodontitis focus in rats and on Cyr61 expression in human bone marrow mesenchymal stem cells(HMSCs).Methods:Rat periapical periodontitis model was established in 32 male SD rats and the rats were divided into normal control group(n=8),model observation group(n=8)and simvastatin treatment group(n=16).The rats were respectively sacrificed at the scheduled times.The area of the periapical lesions was measured by radiographs and HE staining.HMSCs were treated with simvastatin,the activity of alkaline phosphatase(ALP)was observed by ALP staining,the expression of Wnt,β-catenin and Cyr61 were examined by western blot.Results:There was no obvious lesion in normal control group and large lesion appeared in model observation group(P<0.05).The periapical lesion area of the simvastatin treatment group were remarkably smaller than that of the saline group(P<0.01).The activity of ALP and the expression of Wnt,β-catenin and Cyr61 of HMSCs were increased after treated with simvastatin.Conclusion:Simvastatin can promote the healing of bone tissue in the rats with periapical periodontitis and improve osteogenesis by Wnt signaling pathway.
