光交联粘弹性水凝胶快速恢复损伤膝关节运动的实验研究 被引量:2

摘要 目的:构建一种新型水凝胶,快速恢复损伤关节的运动功能。方法:以明胶作为硬链,透明质酸作为软链,通过与甲基丙烯酸酐加成反应,增强两种材料性能与分子间物理交联作用;同时筛选光交联引发剂与配套紫外光源照射参数,最终形成以明胶为网状支架,内部均匀填塞透明质酸的稳定结构,并具有粘弹性承力特性的新型水凝胶。建立20只SD大鼠膝关节损伤模型,分别植入新型水凝胶和对照材料,1周后即进行滚轮运动实验,检测动物运动功能恢复情况,2月后进行病理切片,检测其最终改建效果。结果:该新型水凝胶2 min即可实现光交联固化并具有粘弹性承力特性;与对照组相比,可快速恢复动物损伤关节运动,具有更长的运动时间;2月后材料完全降解,再生为软骨组织。结论:该新型水凝胶极大的缩短了损伤关节运动功能恢复时间,有望在关节镜等技术辅助下实现相关疾病的微创治疗。 Objective:To construct a novel hydrogel for rapid restoration of the motor function of injured joints.Methods:The new hydrogel was designed with gelatin as the stiff chain and hyaluronic acid as the soft chain.Material properties and intermolecular physical crosslinking reaction were enhanced through methacrylic anhydride addition.The photocrosslinking initiator and UV light source irradiation parameters were selected to form a stable structure with gelatin as the mesh scaffold and uniform internal filling with hyaluro-nic acid;the prepared hydrogel had viscoelastic load-bearing properties.Knee joint modle was established in 20 make SD rates.The novel material and the control materials were respectively implanted into the injured joint area,and the animals were tested for the recovery of motor function using the wheel-running exercise 1 week after operation,and the final reconstruction effects were examined 2 months after operation by pathological observation.Results:The novel hydrogel had a stable structure with viscoelastic load-bearing properties after photocross-linke and cured for 2 min.It restored the movement of injured joints in the animals for longer movement duration and distance compared with the control materials.The material degraded completely and regenerated into cartilage tissue 2 months after implantation.Conclusion:The novel hydrogel greatly shortened the motor function recovery time of injured joints and is expected to achieve the minimally invasive treatment of related diseases with the assistance of arthroscopy and other techniques.
