基于小RNA测序的甜樱桃病毒种类检测及鉴定 被引量:2

摘要 病毒病是甜樱桃的重要病害之一,利用高通量测序技术鉴定病毒病害种类,通过对2个表现“花脸”症状的甜樱桃样品YT-1和YT-2进行小RNA测序,并对获取的小RNA进行组装注释。结果表明,样品YT-1获得14190241条reads,鉴定到的主要病毒是樱桃小果病毒2,占小RNA的55.12%,还包括李属坏死环斑病毒、樱桃病毒病A、李树皮坏死茎纹孔伴随病毒、樱桃绿环斑驳病毒及啤酒花矮化类病毒;样品YT-2获得14954849条reads,鉴定到的主要病毒是李属坏死环斑病毒,占小RNA的59.05%,还包括樱桃病毒A、李树皮坏死茎纹孔伴随病毒及樱桃绿环斑驳病毒。 Virus disease is one of the important diseases of sweet cherries,which seriously affects the sustainable development of the sweet cherry industry in Shandong Province.High-throughput sequencing technology played a crucial role in the species identification of virus disease types.The research aimed to sequence small RNA from two samples of sweet cherrys with“dappled”symptoms and then to make assemble and annotations after integrating small RNA samples received.The results showed that the sample YT-1 had obtained 14190241 reads and the primary virus disease identified mainly referred to the Little cherry virus 2(LChV2),accounting for 55.12%of small RNA,which also included Prunus necrotic ringspot virus(PNRSV),Cherry virus A(CVA),Plum bark necrosis stem pitting-associated virus(PBNSPaV),Cherry green ring mottle virus(CGRMV)and Hop stunt viroid(HSVd)23;the samples of YT-2 had obtained 14954849 reads,and the primary virus disease mainly referred to Prunus necrotic ringspot virus(PNRSV),accounting for 59.05%of small RNA,which also included Cherry virus A(CVA),Plum bark necrosis stem pitting-associated virus(PBNSPaV)and Cherry green ring mottle virus(CGRMV).The results can offer theoretical support for prevention and control of viruses occurring in sweet cherry in Shandong Province.
