变应性鼻炎患者外周血PBMC中miR-146a和STAT5b的表达及临床意义 被引量:2

Expression and clinical significance of miR-146a and STAT5b in peripheral blood mononuclear cells of patients with allergic rhinitis

摘要 目的探讨变应性鼻炎患者外周血单个核细胞(PBMC)中微小RNA-146a(miR-146a)、信号转导和转录激活因子5b(STAT5b)的表达及其临床意义。方法选取2017年1月至2020年12月在陕西中医药大学第二附属医院治疗的87例变应性鼻炎患者纳入观察组(其中轻度39例,中重度48例),同时选取在我院治疗的鼻中隔偏曲患者80例作为对照组,检测并比较两组患者PBMC中的miR-146a和STAT5b表达水平,同时采用Pearson法分析miR-146a、STAT5b相对表达量与鼻部症状评分相关性。结果观察组患者PBMC中miR-146a和STAT5b相对表达量分别为2.15±0.98和0.58±0.18,明显低于对照组的3.65±1.00和1.53±0.34,差异均具有统计学意义(P<0.05);观察组中重度患者PBMC中miR-146a和STAT5b相对表达量分别为1.94±0.92和0.41±0.16,明显低于轻度患者的2.41±0.97和0.79±0.20,差异均有统计学意义(P<0.05);观察组患者PBMC中miR-146a和STAT5b相对表达量与鼻部症状评分呈负相关(r=-0.544、-0.577,P<0.05),miR-146a与STAT5b呈正相关(r=0.645,P<0.05)。结论变应性鼻炎患者PBMC中miR-146a和STAT5b表达下调,与患者病情程度呈负相关,同时两者间存在正相关。 Objective To investigate the expression and clinical significance of microRNA-146a(miR-146a),signal transducer and activator of transcription 5b(STAT5b)in peripheral blood mononuclear cells(PBMC)of patients with allergic rhinitis.Methods A total of 87 patients with allergic rhinitis treated in the Second Affiliated Hospital of Shaanxi University of Traditional Chinese Medicine from January 2017 to December 2020 were selected as the observation group(39 cases were mild and 48 cases were moderate and severe),and 80 patients with deviated nasal septum were selected as the control group.The expression levels of miR-146 a and STAT5b in PBMC were detected and compared between the two groups.Pearson method was used to analyze the correlation between the relative expression levels of miR-146 a and STAT5 b and nasal symptom score.Results The relative expression levels of miR-146a and STAT5b in PBMC of the observation group were 2.15±0.98 and 0.58±0.18,respectively,which were significantly lower than 3.65±1.00 and 1.53±0.34 of control group(P<0.05).The relative expression levels of miR-146a and STAT5b in PBMC of patients with moderate to severe degree in the observation group were 1.94±0.92 and 0.41±0.16,respectively,which were significantly lower than 2.41±0.97 and 0.79±0.20 in patients with mild degree(P<0.05).The relative expression levels of miR-146a and STAT5b in PBMC of the observation group were negatively correlated with nasal symptom score(r=-0.544,-0.577,P<0.05),and miR-146a was positively correlated with STAT5b(r=0.645,P<0.05).Conclusion The expression of miR-146a and STAT5b in PBMC of patients with allergic rhinitis was down-regulated,which was negatively correlated with the severity of the disease,and there was a positive correlation between them.
