基于数据库的大黄素协同化疗药物抗肿瘤机制探讨 被引量:2

摘要 目的通过药物靶点数据库对大黄素分子机制进行模拟探讨,为研究大黄素协同抗肿瘤化疗药物提供新的研究策略。方法借助在线数据库资源收集大黄素已知靶点,通过文献及靶点功能数据库筛选具有协同肿瘤化疗药物的潜在靶点,根据靶点构建蛋白-蛋白相互作用(PPI)网络,对潜在靶点KEGG通路分析,明确其发挥协同作用分子机制。结果大黄素发挥协同作用的靶点76个,通过PPI网络分析筛选出中心基因12个,KEGG通路主要集中在肿瘤过程,神经信号通路、急性髓性白血病等。结论通过数据库再分析的方法研究大黄素协同肿瘤化疗药物的潜在靶点和机制,为分子药理机制研究提供新的方向。 Objective To explore the molecular mechanism of emodin based on the drug target database,so as to provide a new research strategy for the study of the synergistic effect of emodin on anti-tumor chemotherapeutic drugs.Methods Known targets of emodin were collected from online database resources,and potential targets that could synergize with anti-tumor chemotherapeutic drugs were screened by retrieving related literature and target function database.A targets-based protein-protein interaction(PPI)network was constructed to analyze the KEGG pathway of potential targets,and to further clarify the mechanism of the molecule that exerts synergistic effects.Results There were 76 targets of emodin exerting synergistic effects,and 12 central genes were screened by PPI network analysis,and the KEGG pathway was mainly concentrated in tumor process,neural signaling pathway,acute myeloid leukemia,etc.Conclusion The database reanalysis-based study on the potential targets and mechanisms of emodin synergized with anti-tumor chemotherapeutic drugs and provides a new direction for the study of molecular pharmacological mechanisms.
