沙棘种子内生细菌对几种植物种子萌发的影响 被引量:1

摘要 目的:为探究沙棘种子内生细菌对兰州北山几种植物种子萌发的影响,促进其种子繁殖。方法:通过Salkowski比色法,从17株沙棘种子内生细菌中筛选出1株产吲哚乙酸(IAA)能力最强的菌株H2B03,采用其上清液和发酵液两种处理对北山植物种子进行萌发试验。结果:菌株H2B03产IAA的量为22.15μg·mL^(-1);在菌株H2B03的上清液和发酵液处理下,臭椿、灌木铁线莲、角茴香、沙枣和曼陀罗种子均不萌发;在上清液处理下,宁夏枸杞、侧柏、鹅绒藤、波斯菊、刺槐和牵牛花种子的萌发受到明显抑制,侧柏种子与对照处理相比发芽率降低了51.6%(P<0.05),而上清液处理对五叶地锦和细叶韭种子的萌发却有一定的促进作用,其萌发率分别提高了10.0%和31.3%;在发酵液处理下,唯有细叶韭有萌发的趋势,其余种子不发芽。结论:沙棘种子内生细菌H2B03菌液可以促进细叶韭和五叶地锦种子的萌发。 Objective:In order to explore the effect of Hippophae rhamnoides seed endophytic bacteria on seed germination of several plants in Beishan,Lanzhou of China.Methods:Through Salkowski colorimetric method,the strain of H2B03 with the strongest ability of producing indoleacetic acid(IAA)was selected from 17 endogenous bacteria of H.rhamnoides seeds,and its supernatant and fermentation broth were used to germinate the seeds of Beishan plant,respectively.Results:The results showed that the IAA yield of strain H2B03 was 22.15μg·mL^(-1);the seeds of Ailanthus altissima Mill.,Clematis fruticosa Turcz,Hypecoum erectum L.,Elaeagnus angustifolia Linn.and Datura stramonium Linn.did not germinate under the treatment of supernatant and fermentation liquid of strain H2B03;the germination of Lycium barbarum L.,Platycladus orientalis(L.)Franco,Cynanchum chinenese R.Br.,Cosmos bipinnata Cav.,Robinia pseudoacacia L.and Pharbitis nil(L.)Choisy seeds were significantly inhibited by the supernatant of strain H2B03.Compared with the control,the germination rate of P.orientalis seeds decreased by 51.6%(P<0.05),while the supernatant treatment promoted the seed germination of Parthenocissus quinquefolia(L.)Planch and Allium tenuissimum L.,and the germination rate increased by 10.0%and 31.3%respectively.Under the treatment of fermentation broth of strain H2B03,only A.tenuissimum had a tendency to germinate,and the other seeds did not germinate.Conclusion:The endophytic bacterial solution H2B03from the seeds of H.rhamnoides could promote the germination of the seeds of P.quinquefolia and A.tenuissimum.
