摘要
目的研究利多卡因对宫颈癌HeLa细胞生物学行为的影响和潜在机制。方法采用四甲基偶氮唑蓝(MTT)法、流式细胞术、Transwell实验分别检测不同浓度的利多卡因对HeLa细胞增殖、迁移、侵袭及凋亡的影响。实时定量PCR(RT-qPCR)检测miR-328表达水平。采用上述方法检测上调miR-328或下调miR-328对HeLa细胞生物学行为的影响。结果利多卡因处理显著降低HeLa细胞增殖率、迁移和侵袭细胞数,增加miR-328表达,并促进细胞凋亡(P<0.05)。上调miR-328表达显著降低HeLa细胞增殖率、迁移和侵袭细胞数,促进细胞凋亡(P<0.05),下调miR-328表达具有相反作用(P<0.05)。上调miR-328表达能够减弱利多卡因对HeLa细胞生物学行为的影响(P<0.05)。结论利多卡因通过上调miR-328表达可抑制宫颈癌细胞HeLa增殖、迁移和侵袭,诱导细胞凋亡。
Objective To study the effect of lidocaine on the biological behavior of HeLa cells in cervical cancer and its underlying mechanism. Methods The effects of lidocaine with different concentrations on proliferation, migration, invasion and apoptosis of HeLa cells were determined by MTT assay, flow cytometry and Transwell assay. Real-time quantitative PCR(RT-qPCR) was applied to detect the expression level of miR-328. The effects of miR-328 up-regulation or down-regulation on the biological behavior of HeLa cells were detected by the above methods. Results Lidocaine treatment significantly reduced the proliferation rate, migration and invasion of HeLa cells, increased miR-328 expression, as well as promoted cell apoptosis(P<0.05). miR-328 up-regulation significantly reduced proliferation rate, migration and invasion of HeLa cells, and promoted apoptosis(P<0.05), while miR-328 down-regulation had the opposite effect(P<0.05). miR-328 up-regulation could reduce the effect of lidocaine on the biological behavior of HeLa cells(P<0.05). Conclusion Lidocaine can inhibit the proliferation, migration and invasion and induce apoptosis in cervical cancer cell HeLa by up-regulating miR-328 expression.
