摘要
目的观察千金消澼液(QXJP)对体外肠上皮炎症损伤模型中紧密连接蛋白(TJ)的影响,探讨其对肠上皮炎症损伤的保护机理。方法利用CCK-8法确定QXJP等药物对于Caco-2细胞的适宜给药浓度;后以脂多糖(LPS)刺激Caco-2细胞制备体外肠上皮炎症损伤模型,分为NC组、LPS组、柳氮磺吡啶(SASP)组和QXJP 1.0、2.5、5.0 mg·mL^(-1)组,预保护给予相应的药物后,以Western Blot法检测Zo-1、Occludin、Claudin-1的表达水平。结果与NC组比较,LPS组Zo-1、Occludin、Claudin-1蛋白水平显著降低(均P<0.05),证明建模成功。与SASP组比较,QJXP 2.5 mg·mL^(-1)组Zo-1、Occludin、Claudin-1蛋白水平无显著差异(P>0.05);QJXP 1.0、5.0 mg·mL^(-1)组Zo-1、Occludin、Claudin-1蛋白水平均显著降低(P<0.05)。结论QXJP 2.5 mg·mL^(-1)能通过提高Zo-1、Occludin、Claudin-1含量进而保护肠黏膜上皮屏障,从而达到治疗UC的目的。
Objective To observe the effect of Qianjin Xiaopi Enema(QXJP)on the tight junction protein(TJ)in an in vitro model of intestinal epithelial inflammatory injury,and to explore its protective mechanisms on it.Methods The CCK-8 method was used to determine the appropriate administration concentration of QXJP and other drugs for Caco-2 cells.After that,Caco-2 cells were stimulated with lipopolysaccharide(LPS)to prepare an in vitro intestinal epithelial inflammation injury model,which was further divided into an NC group,an LPS group,a sulfasalazine(SASP)group and QXJP 1.0,2.5 and 5.0 mg·mL^(-1)groups.The expression levels of Zo-1,Occludin and Claudin-1 were detected by Western Blot after pre-protection with corresponding drugs.Results Compared with the NC group,the protein levels of Zo-1,Occludin and Claudin-1 in the LPS group were significantly decreased(all P<0.05),which proved that the modeling was successful.Compared with the SASP group,there was no significant difference in the protein levels of Zo-1,Occludin and Claudin-1 in the QJXP 2.5 mg/ml group(P>0.05),while the protein levels of Zo-1,Occludin and Claudin-1 in the QJXP 1.0 and 5.0 mg/ml groups were significantly decreased(P<0.05).Conclusion QXJP 2.5 mg·mL^(-1)can protect the intestinal mucosal epithelial barrier by increasing the content of Zo-1,Occludin and Claudin-1,so as to achieve the goal of treating UC.
