不明原因复发性流产患者蜕膜组织中OX40/OX40L信号轴表达与T细胞亚群相关因子的关系研究

The relationship between the expression of OX40/OX40L signal axis in decidual tissue of patients with unexplained recurrent spontaneous abortion and T cell subsets related factors

摘要 目的分析研究OX40/OX40L信号在不明原因复发性流产患者绒毛组织中的表达及其临床意义。方法选取43例不明原因复发性流产患者(URSA组)和40例行人工流产术孕妇(NC组),RT-PCR和Westernblot分析OX40/OX40L-NFAT1信号途径相关基因和蛋白表达情况,流式细胞术检测T细胞亚群变化,分析OX40/OX40L-NFAT1信号途径相关基因与URSA的关系。结果URSA组CD4^(+)T、CD4^(+)/CD8^(+)显著高于NC组,而CD8^(+)T、CD4^(+)CD25^(+)Treg显著低于NC组(P<0.05)。RT-PCR和Western blot显示,URSA组OX40、OX40L、NFAT1 mRNA和蛋白均显著高于NC组(P<0.05或P<0.01)。相关性分析显示,OX40、OX40L、NFAT1与CD4^(+)T、CD8^(+)T、CD4^(+)CD25^(+)Treg存在相关性(P<0.05或P<0.01)。Logistic回归分析显示,CD4^(+)T、CD8^(+)T、CD4^(+)CD25^(+)Treg、TGF-β1、OX40 mRNA、OX40L mRNA及NFAT1mRNA为URSA的影响因素。结论OX40、OX40L、NFAT1基因和蛋白与URSA母胎T细胞亚群相关细胞因子,尤其是Th17/Treg失衡存在一定相关性,OX40、OX40L表达升高可能与URSA患者母胎免疫功能耐受有一定关联性。 Objective To analyze the relationship between the expression of OX40/OX40L signal axis in decidua tissue of patients with unexplained recurrent miscarriage. Methods 43 patients with unexplained recurrent spontaneous abortion(URSA group) and 40 pregnant women with spontaneous abortion(NC group) were enrolled. RT-PCR and Western blot were used to analyze the expression of OX40/OX40 L-NFAT1 signaling pathway-related genes and proteins. The changes of T cell subsets were detected by flow cytometry. And the relationship between OX40/OX40 L-NFAT1 signaling pathway-related genes and URSA was analyzed. Results The CD4^(+)T and CD4^(+)/CD8^(+)in the URSA group were higher than those in the NC group significantly, while the CD8^(+)T and CD4^(+)CD25^(+)Treg were lower than the NC group significantly(P<0.05). RT-PCR and Western blot results showed that OX40, OX40 L, NFAT1 genes and proteins were significantly higher in URSA group than in NC group(P<0.05 or P<0.01). Correlation analysis showed that OX40, OX40 L, NFAT1 were associated with CD4^(+)T, CD8^(+)T,CD4^(+)CD25^(+)Treg(P<0.05 or P<0.01). Logistic regression analysis showed that CD4^(+)T, CD8^(+)T, CD4^(+)CD25^(+)Treg, TGF-β1,OX40 mRNA, OX40 L mRNA and NFAT1 mRNA were the influencing factors of URSA. Conclusion There is a certain correlation between OX40, OX40 L, NFAT1 genes and proteins and URSA. The increasing expression may be related to the maternal fetal immune function tolerance in URSA patients.
