摘要
目的:以“浊瘀”和小肠“泌清别浊”为理论基础,探讨四妙丸对肠道尿酸排泄的影响。方法:55只雄性SD大鼠按随机数字表法分为正常组10只,模型组、苯溴马隆组(4.7 mg/kg)、四秒丸高、中、低剂量组(2260.6 mg/kg、1130.3 mg/kg、565.2 mg/kg)各9只,以氧嗪酸钾和次黄嘌呤制备高尿酸血症(Hyperuricemia,HUA)大鼠模型,连续21 d。第7天验证模型后剔除造模失败大鼠,每组设置8只。除正常组外,第8天开始给药灌胃每天1次,持续14 d。采集血样、尿液、十二指肠液、空肠液、回肠液,测定和计算指标[包括血清尿酸(Serum uric acid,SUA)、尿酸清除率(Clearance of uric acid,CUA)、肠道总尿酸(Total intestinal uric acid,TUA)、十二指肠尿酸(duodenum uric acid,DUA)、空肠尿酸(jejunum uric acid,JUA)、回肠尿酸(Ileum uric acid,IUA)]。结果:模型组大鼠SUA显著升高,CUA显著降低,TUA、DUA、IUA、JUA无明显升高。与模型组比较,苯溴马隆组SUA显著下降,CUA显著升高,TUA、DUA、JUA、IUA无显著差异;四妙丸各剂量组SUA显著降低,四妙丸中剂量组JUA显著升高,四妙丸低剂量组CUA、JUA、IUA显著升高。结论:四妙丸可促进肠道尿酸排泄,降低SUA水平。
Objective:To investigate the effect of the Simiao pill on intestinal uric acid excretion,based on the theory of"turbid blood stasis"and"clearing away turbid blood"in the small intestine.Methods:55 SD male rats were randomly divided into the normal group(n=10),the model,benzbromarone(4.7 mg/kg),and the high-,middle-,low-dose Simiao Pill groups(2260.6 mg/kg,1130.3 mg/kg,565.2 mg/kg,respectively.)(n=9).The Hyperuricema(HUA)model was treated with potassium oxonate and hypoxanthine for 21 consecutive days.After the validation of the model on day 7,rats with model failure were excluded,and 8 rats were set in each group.From the 8th day,rats were intragastrically administered with corresponding medication once daily for 14 days.Blood,urine,and the duodenal,jejunal,ileal juices were collected on day 21.The levels of serum uric acid(SUA),clearance of uric acid(CUA),total intestinal uric acid(TUA),duodenum uric acid(DUA),jejunum uric acid(JUA)and ileum uric acid(IUA)were measured.Results:The SUA was increased(P<0.01)and the CUA was decreased significantly in the model group(P<0.05),but there was no significant difference in TUA,DUA,JUA,and IUA.Compared with the model group,the SUA was decreased significantly(P<0.01),and the CUA was significantly increased(P<0.01)in the benzbromarone group,while the TUA DUA,JUA,and IUA were not significantly different.The SUA of all Simiao Pill groups were decreased significantly compared with the model group,the JUA in middle-dose Simiao Pill group was significantly increased,and the CUA,JUA,and IUA in low-dose Simiao Pill group were significantly increased.Conclusion:Simiao Pill could promote intestinal uric acid excretion to reduce the SUA level significantly.
